Description
GstPrem™ GST-tag Purification Resin, commonly known as GST purification resin, GST purification column, or GST column, is a purification medium used for the simple, fast, and efficient purification of recombinant proteins with GST (Glutathione S-transferase) tags.
GST, which stands for Glutathione S-transferase, is referred to as GST-tagged recombinant protein, GST-tagged protein, or GST-fusion protein when fused with the target protein. GST-tagged recombinant proteins can be selectively bound to GstPrem™ GST-tag Purification Resin, while other proteins cannot be bound. The GST-tagged recombinant proteins can be eluted by an excess solution of reduced glutathione (GSH), thus achieving the separation and purification of GST-tagged proteins. Due to the purification process being conducted under mild non-denaturing conditions, the target proteins obtained can retain their biological activity. Therefore, they can be used for routine structural and functional studies, antibody preparation, protein-protein interactions, protein-nucleic acid interactions, and other research purposes.
This product can be used not only for the purification of GST-tagged recombinant proteins expressed in Escherichia coli but also for those expressed in mammalian cells, insect cells, and baculovirus expression systems.
GST-tagged recombinant proteins exhibit advantages such as high solubility and the ability to maintain the intact activity of the target protein when expressed in Escherichia coli. Many eukaryotic proteins are expressed in the form of inclusion bodies in Escherichia coli, but when they are fused with GST and expressed in Escherichia coli, a considerable portion can achieve soluble expression, which is advantageous for subsequent purification.
If there are site-specific protease recognition sites between the GST tag and the target protein, such as PreScission Protease, TEV Protease, or Thrombin, the corresponding protease can be used to cleave off the GST tag.
Typically, adding a GST tag at the N-terminus of the target protein can retain the enzymatic activity of GST, facilitating the purification of GST-tagged recombinant proteins using GstPrem™ GST-tag Purification Resin.
This product utilizes a highly cross-linked 6% agarose gel as the matrix and employs optimized linker and GSH coupling technologies to achieve high capacity and high specificity binding of GST-tagged recombinant proteins. Compared to most similar products on the market, it has lower non-specific protein binding, greater pressure tolerance, and more stable GSH coupling.
The gel particles in this product have a diameter of 45-165 μm. The maximum tolerable pressure is 0.025 MPa, which is approximately 5.8 psi. When performing protein purification at a fixed flow rate, the recommended flow rate is 0.5 ml/min.
This product is stored in 20% ethanol, with 5 ml of gel and 5 ml of liquid in a total volume of 10 ml. Before use, it is recommended to fully resuspend the gel before aspiration.
This product has a high binding capacity for GST-tagged recombinant proteins, with a maximum binding capacity of 5-6 mg protein/ml gel for GST. This is comparable to similar products from internationally renowned brands. The actual maximum binding capacity during usage depends on the molecular weight of the GST-tagged recombinant protein being purified. The larger the molecular weight, the greater the maximum binding capacity, and vice versa. For GST-tagged recombinant proteins with a molecular weight of 50 kDa, the actual maximum purification capacity per ml of gel is approximately 8-12 mg. For GST-tagged recombinant proteins with a molecular weight of 100 kDa, the actual maximum purification capacity per ml of gel is approximately 12-18 mg. However, for proteins with the same molecular weight, the maximum binding capacity may vary due to differences in protein characteristics.
Precautions
- Please do not store this product at temperatures below -20°C.
- The gel should be kept moist throughout the storage and purification process.

- If centrifugation does not completely remove insoluble materials from the protein sample, the sample solution can be filtered using a 0.45μm filter membrane.
- The purification of GST-tagged proteins should always be conducted under non-denaturing conditions. If the fusion protein is expressed in the form of inclusion bodies, it is necessary to dissolve the inclusion bodies in 8M urea or 6M guanidine hydrochloride, remove urea and guanidine hydrochloride through dialysis, and refold the protein before using this product for purification.
- After collecting protein samples, it is recommended to complete the purification process as soon as possible and store them at 4°C or on ice to slow down protein degradation. To effectively inhibit protein degradation, an appropriate amount of protease inhibitor mixture can be added to the lysis solution.
- This product is intended for scientific research by professionals and should not be used for clinical diagnosis or treatment, food or drug purposes, or stored in ordinary residential areas.
