Description
GoodView™ Green Nucleic Acid Stain is a non-volatile, oil-based macromolecule that does not easily sublimate or penetrate intact cell membranes, reducing potential cytotoxicity. At typical gel staining concentrations, no mutagenic effects have been observed. This stain offers enhanced safety, high sensitivity, and is an ideal nucleic acid dye for electrophoresis applications across various fragment sizes. It is fully compatible with standard gel imaging systems and visible light-excited gel observation devices, making it suitable for UV gel imaging systems and blue-light-excited gel observation instruments.
Features
- Enhanced Safety: Due to its oil-based macromolecular structure, GoodView™ Green does not readily penetrate intact cell membranes, offering safer handling. AMES test results confirm that at working concentrations (used in gel staining), no mutagenicity has been detected, making it an effective substitute for the carcinogenic ethidium bromide (EB) in nucleic acid electrophoresis.

Usage Instructions
I. Agarose Gel Electrophoresis Staining (Recommended Method)
To stain nucleic acids directly within the gel:
- Gel Preparation: Prepare agarose gel using standard procedures. Add 10,000× concentrated SafeGreen, ensuring a final concentration of 1× within the gel (e.g., for 50 mL of gel, add 5 μL of dye). Mix gently and pour the gel.
- Electrophoresis & Observation: Run electrophoresis as usual and analyze results under a suitable imaging system.
II. Immersion Staining Method (Recommended for High-Concentration DNA Recovery)
- Perform electrophoresis using standard procedures. Immersion staining is highly recommended for high-concentration DNA samples requiring gel recovery.
- Prepare staining solution: Dilute 10,000× GoodView™ Green Nucleic Acid Stain stock by approximately 3,300-fold in 0.1M NaCl solution to create 3× staining solution (e.g., add 15 μL of stock solution to 50 mL of 0.1M NaCl).
- Staining procedure: Carefully place the gel in a polypropylene container and slowly add an adequate amount of 3× staining solution to fully submerge the gel. Stain at room temperature with gentle shaking for ~30 minutes. The optimal staining time may vary depending on gel thickness and agarose concentration (for 1% gels, ~30 minutes is recommended).
- Observation: View results using a UV gel imaging system with 302 nm excitation.
⚠ Note: Immersion staining consumes more dye than in-gel staining. 3× GoodView™ Green staining solution can be reused up to three times when stored at room temperature, protected from light.
Storage
Store at 2–8°C, protected from light and in a dry environment. The shelf life is 12 months.
Precautions
- Heat Stability: GoodView™ Green exhibits excellent thermal stability and can be added directly to hot agarose solutions without waiting for cooling. Mix thoroughly via shaking, stirring, or inversion. Alternatively, add the stain to agarose powder and electrophoresis buffer, then heat in a microwave or other heating method to prepare the gel. This dye is compatible with all common electrophoresis buffers.
- Troubleshooting Band Quality: If bands appear diffuse or poorly separated, try immersion staining to verify whether the issue stems from the dye. If staining does not resolve the issue, consider:
- Reducing agarose concentration
- Using a longer gel
- Increasing electrophoresis time for sharper edges
- Refining sample loading techniques or switching to immersion staining
- Material Compatibility: GoodView™ Green binds to glassware and non-polypropylene materials. When diluting, storing, or staining, use polypropylene containers to minimize dye adsorption.
- Polyacrylamide Gel Staining: Use immersion staining for polyacrylamide gels.
