Exonuclease I (E.coli, 3' to 5') exhibits 3´→5´ exonuclease activity, specifically hydrolyzing single-stranded DNA. It is utilized to remove single-stranded primers in PCR reactions prior to Sanger DNA sequencing or SNP analysis, as well as to eliminate single-stranded primers used in nested PCR reactions or linear single-stranded DNA, leaving double-stranded DNA in the sample. Our Exonuclease I is recombinantly expressed and purified through multiple steps from E. coli. It has a molecular weight of 51 kDa.

Description

Exonuclease I (E.coli, 3' to 5') exhibits 3´→5´ exonuclease activity, specifically hydrolyzing single-stranded DNA. It is utilized to remove single-stranded primers in PCR reactions prior to Sanger DNA sequencing or SNP analysis, as well as to eliminate single-stranded primers used in nested PCR reactions or linear single-stranded DNA, leaving double-stranded DNA in the sample. Unit Definition

One unit of the enzyme is defined as the amount required to catalyze the release of 10 nmol of acid-soluble nucleotides within 30 minutes at 37°C.

 

Features

  • ssDNA-specific 3´ exonuclease activity
  • Heat inactivation: 80°C for 20 minutes

 

Applications

  • Removal of 3´ single-stranded DNA while retaining double-stranded DNA (dsDNA)
  • Degradation of primers after PCR amplification
  • Primer degradation following nested PCR amplification
  • Experiments requiring the removal of single-stranded DNA (ssDNA)
