Co-immunoprecipitation (Co-IP) is a commonly used method for studying protein-protein interactions. The basic principle involves using an endogenous target protein within cells as bait. The antibody against the target protein is co-incubated with the total cellular protein to promote the formation of an immune complex. Protein A+G, which can bind to the Fc region of the antibody, is then added, forming a "binding protein-target protein-target protein antibody-protein A+G beads" complex. After purifying this complex, the proteins are separated by gel electrophoresis and the binding proteins of the target protein are identified using Western blot or mass spectrometry techniques.

Storage

Store at -20°C. This kit is valid for one year from the date of purchase.

 

Notes

  • When lysing cells, perform all operations on ice as much as possible to prevent protein degradation.
  • When measuring protein concentration, the presence of EDTA, EGTA, DTT, ammonium sulfate, and lipids in the sample may affect the results.
  • Ensure the specificity of the antibody, meaning that adding the antibody to cell lysates without the antigen should not cause co-precipitation. This ensures that the co-precipitated protein is obtained by the added antibody and not by non-specific external proteins. Using monoclonal antibodies helps to avoid contamination.
  • When centrifuging Protein A+G, avoid excessive centrifugal force as it can damage Protein A+G. A centrifugal force of 3000-5000×g is sufficient.
