Description
Cell Tracking Blue Fluorescence Staining Kit with Blue CMAC is a kit for live cell fluorescent tracking using Blue CMAC as the blue fluorescent probe. This kit is compatible with fluorescence detection systems including fluorescence microscopes, laser scanning confocal microscopes, fluorescence microplate readers, and flow cytometers. The kit supports subsequent cell fixation and permeabilization, and it is also suitable for staining dead cells or pre-fixed cells.
Blue CMAC, full name 7-Amino-4-chloromethylcoumarin, is a chloromethyl derivative of aminocoumarin. Its molecular formula is C₁₀H₈ClNO₂, molecular weight 209.63, and CAS number 147963-22-2. As a cell-membrane-permeable fluorescent probe, Blue CMAC penetrates the cell membrane and enters live cells. It is hydrolyzed by intracellular esterases to release reactive chloromethyl groups with weak fluorescence. The reactive groups then undergo irreversible covalent binding with the thiol groups (-SH) of intracellular reduced glutathione (GSH), forming CMAC-GSH adducts with strong blue fluorescence.
This probe features the following properties:
- It emits bright blue fluorescence upon excitation, with an excitation maximum at approximately 353 nm and an emission maximum at approximately 466 nm.
- Its chloromethyl group reacts irreversibly with intracellular GSH catalyzed by glutathione S-transferase (GST) to form a fluorescent product that is well retained within cells, enabling long-term labeling.
- The fluorescent signal remains highly stable at physiological pH for at least 48 hours, making it ideal for tracking studies of cell motility, localization, proliferation, migration, chemotaxis and invasion.
- Within a cell population, the dye is passed only to daughter cells and does not transfer to neighboring cells.
- It is non-toxic and does not affect cell viability or proliferation.
Storage
Store at -20 ºC; valid for 1 year. Blue CMAC (1000X) must be protected from light.
Precautions
- Repeated freeze-thaw cycles of Blue CMAC (1000X) will compromise detection performance. Aliquot appropriately upon first use to avoid repeated freeze-thawing.
- For trace volumes of liquid, centrifuge for several seconds before each use to fully collect liquid at the bottom of the tube.
- Fluorescent dyes are susceptible to photobleaching. Minimize light exposure to slow down fluorescence quenching.
- When used for long-term cell tracking experiments, Cell-Tracker™ Blue CMAC maintains signals for up to 48 hours. For longer tracking durations, the Cell Tracking Green Fluorescence Staining Kit with Green CMAC or Cell Tracking Red Fluorescence Staining Kit with Red CMAC is recommended, which enable cell tracking for 3–5 days.
- This product is for research use only by professionals. It shall not be used for clinical diagnosis or therapy, food or pharmaceutical applications, and shall not be stored in residential premises.
- For your safety and health, wear a lab coat and disposable gloves during handling.
