AapCas12b protein originates from Alicyclobacillus acidiphilus, an acidophilic bacterium. It is an RNA-guided endonuclease that relies on RNA to mediate nucleic acid cleavage. Under the guidance of a single-stranded guide RNA (sgRNA), Cas12b protein binds to and cleaves specific sites in target DNA. The Cas12b protein exhibits "collateral cleavage" activity when it recognizes and cleaves target double-stranded DNA, efficiently cleaving non-specific single-stranded DNA (ssDNA) in the system. By designing ssDNA probes labeled with fluorescent groups or other markers at both ends, the CRISPR/Cas12b system enables detection and signal amplification of DNA templates, thus achieving target detection. This system has high sensitivity and strong specificity. The lyophilized version of Cas12b can be transported at room temperature, saving the high cost of dry ice transportation.
