Description

BCA Rapid Protein Assay Kit, also known as BCA Rapid Protein Assay Kit with Wide Range or BCA Rapid Protein Assay Kit with Broad Range, is a kit designed for convenient and accurate protein concentration detection within a wide linear range of 0.04-10 mg/mL, requiring only 5 minutes of incubation at room temperature.

This kit is developed based on an improved version of the BCA method, which is one of the most commonly used techniques for protein concentration determination. It provides an enhanced BCA reagent and high-quality protein standards, allowing for simple, convenient, highly stable, sensitive, compatible, and wide-range linear protein concentration measurements with just 5 minutes of room temperature incubation.

The detection principle and method of this kit are essentially consistent with Thermo's Pierce™ Dilution-Free™ Rapid Gold BCA Protein Assay Kit (A55860/A55861/A55862).

Features

  • This kit has an ultra-wide detection range of 0.04-10 mg/ml. The upper detection limit of this kit is 5-10 times that of the BCA Protein Quantification Assay Kit or the Enhanced BCA Protein Assay Kit. This wide concentration range allows for direct protein quantification within the linear detection range without the need for dilution when measuring protein concentrations in routine samples such as cell or tissue lysates.
  • The kit offers high sensitivity and requires a small sample volume. The sample volume for this kit is 1-10 µl, significantly saving sample usage. Under the recommended conditions of 5 minutes incubation at room temperature, the lower detection limit reaches 20 µg/ml, with a minimum detectable protein amount as low as 0.2 µg. Under the conditions of 30 minutes incubation at 37ºC, the lower detection limit can reach 5 µg/ml, with a minimum detectable protein amount as low as 50 ng.
  • The kit provides rapid detection. Under standard detection conditions, absorbance measurement can be performed after 5 minutes of incubation at room temperature.
  • The kit exhibits good linearity within the concentration range of 0.04-10 mg/ml.
  • This kit has good chemical compatibility. When using this kit to measure protein concentration, it is not affected by most chemicals in the samples. However, it is influenced by chelating agents and certain slightly higher concentrations of reducing agents or detergents. Ensure that EDTA does not exceed 5 mM, Tween 20 does not exceed 0.5%, and that there is no dithiothreitol (DTT) or tris(2-carboxyethyl)phosphine hydrochloride (TCEP). This kit is compatible with amine-reactive labeling reagents (such as biotinylation reagents). The detailed compatibility of this kit with various substances in the samples is shown in the table below. If the sample contains 1-2% Tween 20, a blank control containing the corresponding amount of Tween 20 can be set up to perform protein concentration detection. If the sample contains other incompatible chemicals, you can try setting up an appropriate blank control containing the corresponding incompatible compound to confirm whether protein concentration detection can be performed smoothly.

If the concentration of the sample to be tested is particularly low, it is recommended to use the highly sensitive BCA Protein Assay Kit (High Sensitivity). Under the standard detection conditions of 15 minutes incubation at room temperature, the detection limit can reach 2 µg/ml, with a minimum detectable protein amount as low as 40 ng. The sample volume required for testing is 1-20 µl.

 

Storage

Store at 4ºC, valid for one year. BCA Reagent B should be stored protected from light. The Protein Standard (BSA) should be stored as a solution at -20ºC.

 

Precautions

  • It is generally recommended to perform protein concentration detection under the condition of 5 minutes incubation at room temperature. Only when the protein concentration is too low, it is recommended to extend the incubation time and increase the incubation temperature, such as incubating at 37ºC for 30 minutes or longer. Incubation at room temperature for more than 5 minutes may cause the absorbance of high-concentration standards to plateau and not increase further, leading to inaccurate quantification of high-concentration samples.
  • If precipitates form in BCA Reagents A, B, or C after long-term storage, they can be dissolved by heating in a 37ºC water bath or by stirring. If turbidity persists, thoroughly mix and use the solution once it becomes clear.
  • A microplate reader is required, with a measurement wavelength of 480 nm. A 96-well plate is also needed. If a microplate reader is not available, a standard spectrophotometer can be used. In this case, adjust the working solution volume to meet the minimum detection volume of the cuvette, and the sample and standard volumes can be proportionally increased or remain unchanged. When using a spectrophotometer, the number of samples that can be measured per kit may be significantly reduced.
  • If the sample contains EDTA concentrations higher than 5 mM but less than 10 mM, or reducing agents such as dithiothreitol (DTT) at concentrations below 1 mM, or β-mercaptoethanol at concentrations below 0.01%, it is recommended to use the Enhanced BCA Protein Assay Kit.
  • If the sample diluent or lysis buffer has a high background, or if the sample contains EDTA concentrations higher than 10 mM, or high concentrations of reducing agents such as dithiothreitol (DTT) or β-mercaptoethanol, it is recommended to use the Bradford Protein Assay Kit or the Detergent Compatible Bradford Protein Assay Kit.
  • All reagents should be equilibrated to room temperature before use, and stored according to the kit’s requirements immediately after use.
  • This product is for scientific research use only by professionals and should not be used for clinical diagnosis or treatment, food or drug purposes, or stored in ordinary residences.
