Description

Annealing Buffer for DNA Oligos (5X) is a buffer solution that has been extensively tested and proven effective for DNA oligo annealing. This annealing buffer can be used for regular DNA oligo annealing and is particularly suitable for challenging annealing of DNA oligos used in RNAi (also known as siRNA) plasmid construction. DNA oligos used in RNAi plasmid construction often contain approximately 20 complementary sequences, which can easily form hairpin structures, thereby affecting the proper annealing of the two DNA oligos. By using the Annealing Buffer for DNA Oligos (5X) produced by our company, the formation of hairpin structures in DNA oligos can be effectively avoided, and the two oligos can be directly ligated to enzyme-digested and purified plasmids after annealing. Typically, a large number of positive clones can be obtained after transformation.

The operation of this kit is very simple. Simply mix the DNA oligo to be annealed with the Annealing Buffer for DNA Oligos (5X) in a specific ratio and place it in a PCR machine. The annealing process can be completed in approximately 90 minutes.
