A unit is the amount of enzyme required to catalyze the production of 1 nmol of acid-soluble deoxyribonucleotides from double-stranded DNA substrates in a 50μl reaction system at 37℃ within 30 minutes.

- 1 x T5 Exo Buffer: 50 mM KAc, 20 mM Tris-Ac pH 7.9, 10 mM Mg(Ac)2, 1 mM DTT. The enzyme is also active in PCR Buffer.
- The optimal reaction temperature of the enzyme is 37℃, and it also has partial activity at 50℃, so it can be used for Gibson assembly.
