Recombinant Tobacco Etch Virus Protease (TEV Protease), GMP Grade

Tobacco Etch Virus Protease (TEV Protease) is a recombinant protease derived from the Nla protein of Tobacco etch virus (TEV). It specifically recognizes the seven-amino-acid sequence EXXYXQ(G/S) (e.g., Glu-Asn-Leu-Tyr-Phe-Gln-Gly). This product is recombinantly expressed in Escherichia coli and has a molecular weight of 45kDa. The optimal enzymatic reaction occurs within a pH range of 5.5–8.5 and a temperature range of 30–37°C. The protease remains fully active under conditions containing 1mol/L urea or 1–5mmol/L DTT, β-mercaptoethanol, EDTA, PMSF, aprotinin, leupeptin, and other protease inhibitors.

Description

Tobacco Etch Virus Protease (TEV Protease) is a recombinant protease derived from the Nla protein of Tobacco etch virus (TEV). It specifically recognizes the seven-amino-acid sequence EXXYXQ(G/S) (e.g., Glu-Asn-Leu-Tyr-Phe-Gln-Gly). This product is recombinantly expressed in Escherichia coli and has a molecular weight of 45kDa. The optimal enzymatic reaction occurs within a pH range of 5.5–8.5 and a temperature range of 30–37°C. The protease remains fully active under conditions containing 1mol/L urea or 1–5mmol/L DTT, β-mercaptoethanol, EDTA, PMSF, aprotinin, leupeptin, and other protease inhibitors.

Applications 

TEV Protease is used for the removal of fusion tags such as GST, His-tag, or other tags from expressed fusion proteins.

Notes: 

① CoA indicators can include additional tests like microbial limits, host protein residues, or host DNA residues as per customer requirements. 

② Enzyme activity definition: One activity unit (U) is defined as the amount of enzyme required to cleave 3μg of substrate, achieving over 95% cleavage, in TEV Buffer (50mM Tris-HCl, pH 8.0, 0.5mM EDTA, 1mM DTT) at 30°C for 2 hours.
