Description

Universal miRNA Cloning Linker (5' adenylated, 3' blocked) is commonly used for adding adapters to the 3' ends of RNAs or single-stranded DNAs with hydroxyl groups at their 3' ends in cloning, high-throughput sequencing library preparation, or PCR detection.

This product is 5' adenylated (also referred to as 5' pre-adenylated or 5' pre-adenylation) and 3' blocked, making it suitable for adding adapters to the 3' ends of RNAs with hydroxyl groups. RNA ligase can recognize the "activated" 5' adenylated end of this product and covalently link the 5' end of this product to the 3' hydroxyl end of another single-stranded oligonucleotide without the need for ATP. Using this 5' adenylated, 3' blocked product along with RNA ligase allows for the specific covalent linkage of the 5' end of this product to the 3' hydroxyl end of the target sequence without adding ATP. For example, when ligating with miRNA, it effectively prevents miRNA self-ligation, inter-miRNA ligation, self-ligation of this product, or ligation between different chains of this product, resulting in specific linkage between the 5' end of this product and the 3' hydroxyl of miRNA.

The oligonucleotide sequence of this product is: 5´-rAppCTGTAGGCACCATCAAT–NH2-3´. This sequence is identical to the S1315S Universal miRNA Cloning Linker from NEB.

This product is provided as an aqueous solution with a concentration of 40 ng/µl, which is equivalent to 6.9 µM.

A small package of 1 µg of this product typically allows for 8-17 ligation reactions, while a medium package of 5 µg typically allows for 40-85 ligation reactions. The exact number of ligation reactions depends on the volume of the reaction system and the amount of this product used.

 

Application

This product can be used for small RNA cloning and miRNA library construction.

Storage
