Description

TBE is a white quick-dissolve pellet. Each pouch of TBE quick-dissolve pellets can be used to prepare 1 L of 1× TBE buffer, making it easy and convenient to use. The main components of TBE buffer are Tris, boric acid, and EDTA. The concentration of Tris in 1× TBE buffer is 89 mM, the concentration of boric acid is 89 mM, and the concentration of EDTA is 2 mM.

TBE is a commonly used nucleic acid electrophoresis buffer in biology, primarily for DNA agarose gel electrophoresis. TBE has strong buffering capacity and provides better separation results when used for fragments smaller than 1 kb in electrophoresis. It is also suitable for longer electrophoresis durations. However, TBE can cause high electroosmotic flow when used with agarose gels and can reduce the recovery rate of DNA fragments due to the formation of non-covalent complexes with agarose, so it is not recommended for DNA fragment recovery.

Protocol

1. Measure approximately 600 ml of distilled water and add it to a beaker. Place a magnetic stir bar inside the beaker.
2. Position the beaker on a magnetic stirrer and slowly add the entire contents of one pouch of TBE quick-dissolve pellets. Stir the solution until it is completely dissolved.
3. Add distilled water to the TBE solution from step 2, bringing it to a final volume of 1 L, making it 1× concentration.
