Description
SOD belongs to the metalloproteinase family. There are three types of this enzyme based on the type of bound metal ions:
- Copper-Zinc Superoxide Dismutase (Cu-ZnSOD): The most common form, found in the cytoplasm of cells, appears green.
- Manganese Superoxide Dismutase (Mn-SOD): Found in the mitochondria of eukaryotic cells and some prokaryotic cells, appears purple.
- Iron Superoxide Dismutase (Fe-SOD): Found in prokaryotic cells, appears yellow-brown.
All three types of SOD catalyze the dismutation of superoxide anion radicals into hydrogen peroxide and oxygen. The catalytic reaction can be represented as: 2O2- + 2H+ → H2O2 + O2. Although hydrogen peroxide is still a harmful reactive oxygen species in the body, it is immediately decomposed into harmless water by catalase (CAT) and peroxidases (POD).
Porcine SOD is a copper-zinc superoxide dismutase with a molecular weight of 32,000 Da. It consists of four non-covalently bound homologous subunits, forming a dimer. Each subunit contains 152 amino acids, with a molecular weight of 16,000 Da, and has one disulfide bond and a free thiol group.
Unit Definition
Under constant temperature conditions of 25°C, one enzyme unit is defined as the amount of enzyme that inhibits the autoxidation rate of pyrogallol by 50% per minute in the reaction solution.
Extinction Coefficient: 1 mM = 10.3 (258 nm)
Due to the absence of tryptophan, there is no significant absorption peak at 280 nm.
Optimal pH: 7.8
Optimal Temperature: 25°C
Isoelectric Point: 4.95
Inhibitors: Cyanide, OH- (competitive), hydrogen peroxide
Preparation Instructions
SOD lyophilized powder is light blue-green. It can be reconstituted in 0.1 M potassium phosphate buffer (pH 7.5) to prepare a 1 mg/mL solution and stored in aliquots.
Storage
The lyophilized powder can be stored for over three years at -20°C, one year at 4°C, and six months at 25°C.
A 1 mg/mL solution in 0.1 M potassium phosphate buffer (pH 7.5) remains active for one hour at 60°C, six hours at 25°C, and two days at 4°C. It can be stored for over a year at -20°C.
