Description
Serum-free Cell Freezing Medium is a ready-to-use cell freezing solution that does not contain serum or proteins. It allows for long-term storage of cells at -80°C without the need for programmed cooling and is suitable for the cryopreservation of most mammalian cells. This serum-free cell freezing medium does not contain commonly used fetal bovine serum (FBS) or any animal-origin proteins (animal origin-free, AOF), reducing the risk of cell contamination and ensuring cell safety. Additionally, cells can be directly stored at -80°C using this serum-free cell freezing medium without the need for programmed cooling, making it very convenient to use.
Cell freezing medium, also known as cell culture freezing medium or cell cryopreservation medium, is referred to in English as Cell Culture Freezing Medium, Cell Freezing Solution, or Cell Cryopreservation Medium.
Cell cryopreservation is one of the main methods for cell preservation. By using cryopreservation technology to store cells at -196°C in liquid nitrogen, cells can temporarily exit the growth state and maintain their characteristics for long-term storage. When needed, the cells can be thawed and used for experiments. Additionally, appropriately cryopreserving a certain amount of cells can prevent the inability to continue culturing cells due to contamination or other unexpected events, thus serving as a means of cell preservation.
Features
- This product is a ready-to-use solution, convenient to use. It is a sterile, ready-to-use solution. After collecting the cell pellet to be frozen, add an appropriate amount of freezing medium and mix well. There is no need to prepare any solution yourself, making it very convenient to use.
- This product does not require programmed cooling and can be directly frozen at -80°C. Generally, cell freezing and thawing use the “slow freezing and rapid thawing” method. During the cell freezing process, it is usually necessary to use a programmed cooling box such as the Cell Freezing Box or take programmed cooling steps. However, using this serum-free cell freezing medium, there is no need for programmed cooling, and it can be directly frozen at -80°C.
- This product can store cells long-term at -80°C without using liquid nitrogen. Cells can be directly stored long-term at -80°C using this serum-free cell freezing medium. Alternatively, after placing at -80°C for at least 24 hours, cells can be transferred to liquid nitrogen for storage without the need for programmed cooling steps or using a programmed cooling box, making it very convenient to use.
- This product has a defined chemical composition, is serum-free, and safe to use. It uses a formula with defined chemical components, containing amino acids, vitamins, inorganic salts, glucose, DMSO, and various essential nutrients and cryoprotectants for cells. It contains phenol red but does not contain commonly used fetal bovine serum or any animal-derived proteins, reducing the variability in freezing effects caused by serum batches. It also lowers the risk of contamination by various viruses, fungi, and mycoplasma, ensuring cell safety, and reduces the impact of exogenous proteins on normal cell growth and differentiation.
- Cells frozen with this product have a high recovery rate. The formula of this product has been optimized, and cells frozen with this product have a high recovery rate and maintain good cell viability after thawing. Tests have shown that for cells with a high recovery rate after freezing and thawing, such as HeLa (human cervical cancer cells) and BEAS-2B (human normal lung epithelial cells) , the recovery rate is above 90% after one year of freezing at -80°C. For cells with a lower recovery rate after freezing and thawing, such as Jurkat (human T lymphocyte leukemia cells) and L929 (mouse fibroblasts), the recovery rate is above 65% after one year of freezing at -80°C, which is comparable to the effect of using FBS-containing freezing medium. Compared to similar serum-free cell freezing media, the effect is comparable or better. For some cells frozen with this product, even after being frozen at -20°C for 6 months, the recovery rate is above 50%.
Storage
Store at 4°C, valid for one year. Do not freeze.
Precautions
- Do not store this product at -20°C before freezing cells, as it may cause slight precipitation after thawing. However, tests have shown that even if stored at -20°C and precipitation occurs, it does not significantly affect the freezing effect.
- For certain special or particularly valuable cells, it is recommended to perform comparative tests using conventional FBS-containing freezing medium to confirm the recovery rate before using this product.
- This product contains DMSO. For cells sensitive to DMSO, it is recommended to conduct pre-experiments for freezing.
- For common cells that are not particularly valuable, it is recommended to use this serum-free cell freezing medium for direct long-term storage at -80°C, as it is more convenient to use and operate. However, for more valuable cells or cells with lower recovery efficiency, if better freezing and recovery effects are desired, it is recommended to use the Cell Freezing Box for programmed cooling and transfer to liquid nitrogen for storage after placing at -80°C for at least 24 hours.
- This product uses a bicarbonate buffer system commonly used in cell culture. If exposed to air for too long, it may lead to the release of carbon dioxide, causing the pH to gradually rise and even change the color of the solution. Due to the presence of calcium and magnesium ions in the solution, precipitation of calcium carbonate, calcium hydroxide, and magnesium hydroxide may occur at high pH. During use, minimize exposure time and tightly cap the container after use. If necessary, aliquot the solution to avoid pH increase. If slight precipitation occurs after some time, tests have shown it does not affect the freezing effect.
- Ensure that cells are in good growth condition before freezing, with a viability rate typically greater than 90%.
- After adding the cell freezing medium and mixing well, promptly transfer to a -80°C ultra-low temperature freezer for storage. When storing in a -80°C ultra-low temperature freezer, place the frozen cells inside the freezer to avoid frequent opening of the door, which can cause temperature changes at the door and affect the freezing effect.
- Use an alcohol-resistant marker pen to label, to prevent the markings from being erased by alcohol or isopropanol, making them unrecognizable.
- This product is for scientific research use by professionals only and is not intended for clinical diagnosis or treatment, food or drug use, or storage in ordinary residences.
