Description
DNA restriction enzymes have been widely used in molecular biology for nearly fifty years. However, RNA restriction enzymes have remained largely unexplored. Existing RNA analysis methods are complex, costly, and unstable, often failing to meet the demands of modern molecular biology.
RRE1 functions similarly to EcoToxN1 and belongs to the Type III toxin-antitoxin family. It specifically recognizes and cleaves the pentamer sequence (GAA↓AU) in single-stranded RNA. Unlike conventional RNA cleavage methods, RRE1 does not require guide RNA or DNA pairing, yet it maintains high specificity in target sequence cleavage. Moreover, it remains effective across a broad temperature range, making it an ideal RNA restriction enzyme for RNA studies and biosynthesis applications.

One unit is defined as the amount of enzyme required to cleave 10 pmol of single-stranded RNA substrate containing rGrArArArU in a 15-minute reaction at 25°C.
