Description
Rapid Running Buffer (hereafter referred to as RRB) is a white quick-dissolve pellet. Each pouch of RRB quick-dissolve pellets can be used to prepare 1 L of 1× RRB buffer, making it easy and convenient to use.
RRB buffer is designed for rapid nucleic acid gel electrophoresis, primarily for DNA agarose gel electrophoresis. RRB buffer is characterized by strong buffering capacity and high resolution, effectively separating DNA fragments of up to 5 kb. When used with an appropriate concentration of agarose gel, it supports high-voltage rapid electrophoresis at 300-350 V (15 V/cm), reducing the conventional electrophoresis time from approximately 30 minutes to around 10 minutes. This buffer can serve as a replacement for TBE electrophoresis buffer and is suitable for small molecular weight DNA agarose gel electrophoresis.

Protocol
1. Preparation of 1 L of 1× RRB Buffer
① Measure approximately 600 ml of distilled water and add it to a beaker. Place a magnetic stir bar inside the beaker.
② Position the beaker on a magnetic stirrer and slowly add the entire contents of one pouch of RRB quick-dissolve pellets. Stir the solution until it is completely dissolved.
③ Add distilled water to the RRB solution from step ②, bringing it to a final volume of 1 L, making it 1×.
2. Electrophoresis
① Rapid Electrophoresis It is recommended to use a 1.5% agarose gel, and adjust the voltage to 300-350 V (15 V/cm). Effective separation of DNA fragments ranging from 100 to 5000 bp can be completed within 10 minutes.
Note 1: When performing rapid electrophoresis, it is recommended to use a 1.5% agarose gel. Lower gel concentrations may result in poor resolution for fragments below 200 bp, while higher concentrations may lead to poor resolution for larger fragments.
Note 2: If the room temperature is too high or continuous electrophoresis is performed, it can lead to an elevated electrophoresis buffer temperature. It is recommended to cool down before conducting electrophoresis.
② Standard Electrophoresis 1× RRB electrophoresis buffer can replace TBE for standard electrophoresis procedures, effectively separating small molecular weight DNA fragments.
