Description
RNAFortify is a non-toxic, directly usable sample storage solution that separates RNA from RNAases within cells, allowing for the rapid and reliable preservation of RNA in animal tissues and cells. After tissue collection, immerse it immediately in RNAFortify, and it can be stored at room temperature for 7 days, at 4°C for 4 weeks, and long-term at -20°C or -80°C. The RNA remains stable without degradation, and high-quality RNA can be obtained by extraction using various methods after retrieval.
Protocol
- Calculate the required amount of RNAFortify based on the volume of each sample to be preserved. The amount of RNAFortify should be 10 times the volume of the tissue (e.g., use 1ml RNAFortify for approximately 100mg of tissue); for the collection of 2x10^6 cells, the amount of RNAFortify is 1ml.
- Dispense the required amount of RNAFortify into prepared storage tubes.
- Quickly cut larger tissues into the desired thickness.
- When storing, immerse the sample in RNAFortify and leave it overnight in a 4°C refrigerator (overnight at 4°C is necessary to allow RNAFortify to fully penetrate the tissue). Then transfer to a -20°C freezer (RNAFortify remains liquid at -20°C with crystallization, which is normal); or after overnight storage at 4°C, remove tissue blocks from RNAFortify and transfer to a -80°C freezer. Specimens stored in RNAFortify can undergo repeated freeze-thaw cycles up to 20 times without affecting the quality of RNA.
Precautions
- Tissue and cell collection should be done quickly, and after collection, immerse them in RNAFortify as soon as possible to prevent RNA degradation.
- Frozen tissues cannot be preserved with RNAFortify as it cannot effectively penetrate frozen tissues.
- RNA extraction from stored samples: After removing the sample from a -20°C or -80°C freezer, resuspend the sample at room temperature, remove tissue blocks, and use for RNA extraction. For cell samples, after thawing, centrifuge at low speed to collect cells, remove RNAFortify, and then use for RNA extraction. Subsequent processing (such as tissue homogenization) can be performed at room temperature, and there is no need for operations in liquid nitrogen, as RNA remains effectively protected. Residual traces of RNAFortify preservation solution do not affect the quality of subsequent RNA extraction.
