Description
Aprotinin is a competitive serine protease inhibitor that can inhibit the activity of trypsin, chymotrypsin, and kallikrein. Aprotinin forms a stable complex with proteases, thereby blocking the active site of the enzyme. This binding is reversible, and most aprotinin-protease complexes can be dissociated under extreme pH conditions (>10 or <3.0). Aprotinin binds to proteases on a molar basis. Structurally, aprotinin is a monomeric globular protein derived from bovine lungs, composed of 58 amino acids arranged in a single peptide chain with three disulfide bonds. Recombinant aprotinin is expressed in Escherichia coli, purified by multiple column chromatography, and has an amino acid sequence identical to aprotinin derived from bovine lungs. Recombinant aprotinin possesses the same enzymatic properties as the animal-derived aprotinin and can be used as a substitute for animal-derived aprotinin in various biotechnological processes.
