Description
PVDF Membrane refers to pre-cut or roll-format polyvinylidene fluoride (PVDF) blotting membranes. It is most widely applied for protein transfer from gels to membranes in immunoblotting (Western blot). Additional applications include adsorption assays, amino acid analysis, N-terminal protein sequencing, dot and slot blot assays, glycoprotein staining, and lipopolysaccharide analysis.
This PVDF membrane portfolio is available in two pre-cut formats and a bulk roll. The pre-cut sizes are 6.6 × 8.5 cm and 8.5 × 13.2 cm respectively. The 6.6 × 8.5 cm sheet fits standard full-size gels and eliminates manual trimming for streamlined workflows. The 8.5 × 13.2 cm sheet can be further cut to custom dimensions as needed. The bulk roll measures 26.5 cm × 3.75 m and requires self-trimming to achieve membrane sizes tailored to experimental requirements.
Two pore sizes are offered: 0.22 μm and 0.45 μm, compatible with the majority of immunoblot workflows. The 0.22 μm PVDF membrane is optimized for proteins smaller than 20 kDa, effectively minimizing small-molecular-weight protein leakage while delivering superior protein binding capacity and higher yields for protein sequencing relative to the 0.45 μm variant. The 0.45 μm PVDF membrane serves as the standard choice for routine Western blotting, ideal for proteins larger than 20 kDa.
Following electrophoresis, stain-free SDS-PAGE gels must first be activated under UV light prior to standard protein transfer. After transfer, image capture is performed under UV excitation. Conventional PVDF or nitrocellulose membranes generate intrinsic autofluorescence under UV irradiation, which creates high background noise. Although stain-free protein bands remain visible, resolution and contrast are markedly reduced compared to gel-based imaging.
Our low-background-fluorescence PVDF membranes drastically reduce intrinsic autofluorescence to produce high-quality fluorescent images. This specially optimized membrane exhibits minimal autofluorescence across a broad excitation/emission wavelength spectrum, paired with robust protein binding capacity and exceptional detection sensitivity. Longer exposure times can be applied without compromising signal-to-background ratio, maximizing the detection performance of fluorescent imaging platforms. Designed specifically for fluorescent detection systems (e.g., LI-COR Odyssey) and stain-free SDS-PAGE workflows, it is an ideal substrate for multiplex immunofluorescence detection and quantitative analysis.
PVDF membranes present multiple benefits compared to nitrocellulose membranes. They possess exceptional mechanical strength, resisting tearing and curling, alongside low non-specific background, broad solvent compatibility, and outstanding staining performance. Furthermore, properly processed PVDF membranes support sequential re-probing for multiple target proteins.
Due to the strong hydrophobicity of this PVDF membrane series, pre-soaking in appropriate solvents such as ethanol is required for activation before use, which facilitates efficient protein binding to the membrane.
This PVDF membrane series is compatible with a wide range of staining reagents, including Ponceau S, Coomassie Brilliant Blue, Sypro Ruby, transillumination staining, Toluidine Blue, CPTS, colloidal gold, India ink and Amido Black. Stains such as Ponceau S and CPTS can be completely washed off after visualization, allowing the membrane to be reused for subsequent downstream analysis. In contrast, Coomassie Brilliant Blue, India ink and Amido Black produce irreversible staining, rendering the membrane unsuitable for further experimentation post-staining.
The membranes are optimized for chemiluminescence detection and also compatible with conventional colorimetric staining and radioisotope-based detection workflows.
Storage Conditions
Store at room temperature with a shelf life of 5 years.
Precautions
- Use flat-tipped forceps to handle PVDF membranes, and only grip the membrane edges gently. Avoid scratching, folding or creasing the membrane surface during all operations.
- Owing to high hydrophobicity, the membrane must be activated by soaking in ethanol, methanol or other suitable solvents prior to use. Beyotime’s non-toxic PVDF Membrane Wetting & Activation Solution is recommended. A translucent appearance indicates full activation; if the membrane remains opaque white, continue soaking until fully wetted.
- Once wetted and activated, the membrane must remain hydrated and can be stored in Western transfer buffer or wash buffer. If the membrane dries out accidentally, repeat the full wetting and activation procedure.
- For optimal performance, slowly lower the membrane along the liquid surface during activation to ensure uniform coverage by the wetting solution without trapped air bubbles.
- For short-term re-probing of membranes after chemiluminescence exposure: rinse the membrane with distilled water first, then strip bound antibodies using a suitable Western blot stripping buffer. If antibody stripping cannot be performed immediately, submerge the membrane in TBSTw inside a sealed sampling bag to prevent desiccation, and store the sealed bag at 4 °C for up to one week.
