Description
Traditional microRNA extraction kits are typically based on the principle of using guanidinium thiocyanate/phenol/chloroform (TRIzol method) combined with high-concentration ethanol to adsorb small fragments of microRNA onto a silica membrane. However, complex plant species such as cotton, grapes, poplar, holly, roses, dendrobium, ginseng, rice seeds, ginseng, corn germ, and many other samples are challenging, if not impossible, to extract total RNA that meets the necessary requirements using the TRIzol principle, let alone microRNA. Consequently, most companies that utilize the TRIzol method principle for their microRNA extraction kits, are left helpless when faced with complex plant samples. In fact, their product lines don't even include microRNA extraction kits specifically for complex plants. Users are reluctantly forced to resort to traditional methods or use the TRIzol method and isopropanol/ethanol precipitation to extract microRNA. While this might yield some microRNA, the precipitation method results in significant loss or co-precipitation with impurities, compromising experimental results. Consequently, submitting such findings to international journals often faces scrutiny and can even lead to the retraction of the article.
Our kit, based on the company's global-leading experience in extracting from hundreds of complex plants, innovatively adopts a technical route that does not use phenol or chloroform. We can extract microRNA from the majority of complex plants like cotton, poplar, holly, roses, salvia, etc. Moreover, our kit is also compatible with simpler samples like Arabidopsis, rice, wheat, tobacco, and various other plants.
The unique lysis solution rapidly lyses cells and inactivates cellular RNA enzymes. The plant RNA assist agent, PLANTaid, helps bind polysaccharides and polyphenols and removes them via centrifugation. The lysate then passes through a genomic DNA removal column, wherein the genomic DNA is cleared, and RNA (including microRNA) is selectively filtered. After adjusting the filtered RNA with ethanol, the RNA selectively adsorbs to the silica membrane inside the centrifuge column under high salt conditions. Following a series of quick washes with specialized solutions and centrifugation, cellular metabolites and proteins are removed. Finally, low-salt RNase-free H2O elutes the pure RNA (including microRNA) from the silica membrane.
By using separate extraction steps, it's also possible to individually purify enriched microRNA components and total RNA (>200 nt), thereby obtaining separated enriched microRNA and RNA.
