Description
Mouse Peripheral Blood Lymphocyte Separation Kit is a separation reagent kit based on the density gradient centrifugation principle. It allows for the simple, fast, and efficient separation of lymphocytes and mononuclear cells from mouse peripheral blood, cord blood, or bone marrow cells. The main component of this kit is the Mouse Peripheral Blood Lymphocyte Separation Medium, also known as Lymphocyte Separation Medium (LSM), lymphocyte gradient separation medium, density gradient medium, PBMC separation medium, PBMC sample density separation medium, or PBMC cell separation medium. The Rat Peripheral Blood Lymphocyte Separation Medium in this kit is iso-osmotic with mouse peripheral whole blood.
The density of lymphocytes in rodents is slightly higher than that of human lymphocytes. The Mouse Peripheral Blood Lymphocyte Separation Medium provided in this kit has been optimized and improved to be iso-osmotic with mouse peripheral blood. By using the separation medium in this kit for density gradient centrifugation, the cells or liquid in the centrifuge tube form four layers from top to bottom: the lower-density plasma and platelets, which float on top of the separation medium as the first layer; the slightly lower-density mononuclear cells (PBMC, including lymphocytes and monocytes) located above the separation medium interface, usually forming a ring-like milky layer as the second layer. Therefore, removing the first layer allows for the collection of PBMC. The third layer is a transparent separation medium, and the fourth layer consists of higher-density red blood cells and granulocytes, which settle at the bottom.

Features
- The operation of this kit is simple, with a short processing time and a high separation rate. The entire experiment only requires simple pipetting and centrifugation steps, allowing for the separation of lymphocytes from the sample to be completed in approximately one hour. The extraction efficiency of lymphocyte separation using this kit is greater than 80%. For obtaining target cells with high purity, immunomagnetic beads can be used in combination. By using this kit for pre-separation, the amount of magnetic beads required can be reduced, thus lowering costs. The kit also includes sterile siliconized centrifuge tubes (10ml), sample dilution solution, and cleaning solution, making it more convenient to use.
- This kit is of high quality and undergoes strict quality control. The separation solution in this kit is formulated using pharmaceutical-grade raw materials, optimized, and processed under sterile conditions, with endotoxin levels below 0.5 EU/ml.
- The kit has a wide range of applications. It is suitable not only for diluted anticoagulated blood samples but also for undiluted anticoagulated blood samples. It can separate lymphocytes and also obtain plasma of certain purity. The separated lymphocytes can be directly analyzed or used for cell culture.
Additional Notes
- Mouse Peripheral Blood Lymphocyte Separation Medium should be inverted and thoroughly mixed prior to use to ensure proper blending. It should be aspirated under sterile conditions to prevent any microbial contamination.
- If further culturing of the isolated lymphocytes is desired, it is essential to employ sterile techniques during both the blood collection and separation processes to avoid any microbial contamination.
- The reagent kit should be stored at room temperature and should not be refrigerated or frozen. Exposure to low temperatures may lead to the formation of white crystals in the rat peripheral blood lymphocyte separation fluid, thereby affecting its separation effectiveness.
- Due to the variation in density of the mouse peripheral blood lymphocyte separation fluid at different temperatures (higher density at low temperatures and lower density at high temperatures), it is recommended to use the reagent kit within a temperature range of 18-22ºC, with the optimal separation temperature being 20ºC. It is important to note that different blood samples may yield varying separation results. Factors such as prolonged storage, transportation, the physical condition of the blood donor, medication usage, underlying diseases, and gender can all influence the effectiveness of the separation process.
- Certain types of plastic centrifuge tubes (such as polystyrene, PS) can generate static electricity, leading to cell adhesion and subsequently affecting the separation effectiveness. It is advisable to use sterile siliconized centrifuge tubes or centrifuge tubes made of polypropylene material.
- To ensure optimal lymphocyte separation and prevent a decrease in cell viability, it is recommended to dilute the blood samples with sample dilution fluid, buffer solutions lacking calcium and magnesium ions, or physiological saline.
- Please note that this product is exclusively designed for the separation of peripheral blood lymphocytes in mice and is not suitable for other animal species. Specific separation fluids should be used for blood samples obtained from different animal species.
- This product is solely intended for scientific research purposes and should be handled by professionals only. It should not be used for clinical diagnosis or treatment, food or drug purposes, nor should it be stored in residential areas.

Storage
Please store this product at room temperature, away from direct light. It has a shelf life of two years. It is important to note that refrigeration or freezing of this product is strictly prohibited.
