Description
Human Peripheral Blood Lymphocyte Separation Medium is a separation solution based on the principle of density gradient centrifugation. It allows for the simple, rapid, and efficient isolation of lymphocytes, monocytes, and other cells from human peripheral blood, umbilical cord blood, or bone marrow cells. It is also known as Lymphocyte Separation Medium (LSM), lymphocyte gradient separation medium, density gradient medium, PBMC separation medium, PBMC sample density separation medium, or PBMC cell separation medium. The density of this separation medium is 1.077 g/ml at 20ºC, which is nearly iso-osmotic with whole peripheral blood.
Different blood cells in human peripheral blood vary in terms of volume, morphology, and specific gravity (density). Red blood cells and granulocytes have a higher specific gravity, around 1.092, while mononuclear cells (lymphocytes and monocytes) have a specific gravity of approximately 1.075, and platelets have a specific gravity of about 1.030. Therefore, by using a separation solution with a density ranging from 1.075 to 1.090, lymphocytes and monocytes, which have lower specific gravity, float in this separation medium, while red blood cells and granulocytes, with higher specific gravity, sink. This allows for the isolation of mononuclear cells from peripheral blood cells. The most commonly used separation solution is a mixture with a density of approximately 1.070, prepared using polysucrose 400/Ficoll 400 and sodium diatrizoate.
The human peripheral blood lymphocyte separation medium we provide has undergone formulation optimization and improvement. At 20ºC, its density is approximately 1.077 g/ml, which is nearly iso-osmotic with peripheral blood. During density gradient centrifugation using this separation medium, the cells or liquid in the centrifuge tube separate into four layers from top to bottom: the plasma and low-density platelets form the first layer, suspended in the upper part of the separation medium; the mononuclear cells (PBMC, including lymphocytes and monocytes) have a slightly lower density than the separation medium and are located above the interface of the separation medium. They usually appear as a circular milky-white layer, which is the second layer. By removing the first layer, PBMC can be obtained. The third layer is a transparent separation medium layer, and the red blood cells and granulocytes, with higher density, settle at the bottom, forming the fourth layer.

- This product offers a user-friendly experience with easy operation, a short processing time, and a high rate of lymphocyte separation. The entire experiment can be completed in approximately one hour by following simple pipetting and centrifugation steps. The product achieves an extraction efficiency of over 80% for lymphocyte separation. To obtain target cells of high purity, immunomagnetic beads can be used in combination. By utilizing this reagent kit for pre-separation, the required amount of magnetic beads can be reduced, resulting in cost savings.
- This product maintains a high standard of quality through stringent quality control measures. The separation solution is formulated using pharmaceutical-grade raw materials and optimized for optimal performance. The manufacturing process takes place under sterile conditions, ensuring a sterile product with endotoxin levels below 0.5 EU/ml.
- This product has a wide range of applications and is suitable for both diluted and undiluted anticoagulated blood samples. It effectively separates lymphocytes and can also obtain plasma of a certain purity. The isolated lymphocytes can be directly analyzed or utilized for cell culture purposes.
Additional Notes
- Before using this product, it should be inverted and mixed thoroughly. The aspiration should be performed under sterile conditions to avoid microbial contamination.
- If further cultivation of the separated lymphocytes is desired, sterile techniques should be followed during blood collection and separation to prevent microbial contamination.
- This product should be stored at room temperature and strictly prohibited from refrigeration or freezing. Under low-temperature conditions, the product may form white crystals, which can affect the separation effectiveness.
- Due to the increase in density at low temperatures and decrease at high temperatures, the recommended temperature range for using this product is 18-22ºC, with the optimal separation temperature being 20ºC.
- Different blood samples may yield varying separation results. Umbilical cord blood, for example, may exhibit suboptimal separation efficiency due to factors such as prolonged ex vivo time and transportation. The host's physical condition, medication usage, underlying health conditions, and gender can also influence the separation effectiveness.
- Certain types of centrifuge tubes made of plastics such as polystyrene (PS) may generate static electricity, leading to cell adhesion and affecting separation results. It is recommended to use polypropylene centrifuge tubes.
- To minimize the reduction in cell viability and improve lymphocyte separation efficiency, it is advisable to dilute blood samples using buffer solutions or physiological saline that do not contain calcium or magnesium ions.
- Due to the density differences of immune cells among different animal species, this product is only suitable for the separation of lymphocytes from human peripheral blood and is not applicable to other animals. Specific separation solutions should be used for blood samples from different animal species.
- This product is intended for scientific research purposes by professionals only and should not be used for clinical diagnosis or treatment, or for food or drug applications. It should not be stored in residential areas.

Storage
Please store this product at room temperature, away from direct light. It has a shelf life of two years. It is important to note that refrigeration or freezing of this product is strictly prohibited.
