Description

Membrane and Cytosol Protein Extraction Kit provides a simple and convenient method for extracting membrane proteins and cytosolic proteins from cultured cells or tissues. The extracted membrane proteins include not only those from the plasma membrane but also from mitochondrial membranes, endoplasmic reticulum membranes, and Golgi membranes.

This kit works by homogenizing cells to moderately break them apart, followed by low-speed centrifugation to remove the nuclear fraction and any remaining intact cells. The supernatant is then subjected to high-speed centrifugation to obtain the membrane pellet and the supernatant containing cytosolic proteins. Optimized membrane protein extraction reagents are used to extract membrane proteins from the pellet.

The separation and extraction of membrane proteins and cytosolic proteins from cultured cells or tissues can be completed in approximately 90 minutes. The extracted proteins can be used for subsequent experiments such as SDS-PAGE, Western blotting, and enzyme activity assays.

The membrane protein extraction reagents contain protease inhibitors, phosphatase inhibitors, and EDTA, making them unsuitable for enzyme activity assays involving proteases or phosphatases affected by these inhibitors. However, the extracted membrane or cytosolic proteins are suitable for detecting protein phosphorylation levels.

Following the instructions provided in the manual, this kit can extract proteins from 100 cell or tissue samples.

Store at -20°C, valid for one year.

Precautions

• PMSF (phenylmethylsulfonyl fluoride) is required. Add PMSF to the extraction reagent 2-3 minutes before adding it to the sample to prevent PMSF from quickly losing its effectiveness in aqueous solution. PMSF can be ordered from us.
• The membrane and cytosolic proteins extracted using this kit can be directly quantified using our BCA Protein Assay Kit. The extracted membrane proteins are not suitable for protein concentration determination using the Bradford method.
• This product is for scientific research use only by professionals and is not intended for clinical diagnosis or treatment, food, or drugs, and should not be stored in ordinary residences.
