In Vitro Study

Magnolin, a natural compound abundant in Magnolia flos, has long been utilized in oriental medicine to alleviate headaches, nasal congestion, and inflammatory reactions. It specifically targets the active pockets of ERK1 and ERK2, pivotal signaling molecules in cancer cell metastasis. By suppressing the ERKs/RSK2 signaling pathway, Magnolin effectively inhibits NF-κB transactivation activity. Additionally, it curtails the production of tumor necrosis factor-α (TNF-α) and prostaglandin E2 (PGE2) through the inhibition of extracellular signal-regulated kinases (ERKs), crucial in regulating cell proliferation, transformation, and cancer cell metastasis. Notably, Magnolin treatment significantly suppresses JB6 Cl41 cell migration induced by EGF treatment in a dose-dependent manner. Furthermore, Magnolin inhibits ERK1/2/RSK2 signaling-mediated IκBα phosphorylation at Ser32, thereby inhibiting NF-κB activation and subsequent cell migration.

Storage

Store at -20°C for at least one year. The solid powder is stable at 4°C for at least one month. If dissolved in a non-DMSO solvent, it is recommended to aliquot and store at -80°C for up to six months.
