Description
Lipid Droplet Red Fluorescence Assay Kit with LD540, also known as the LD540 Staining Kit or Red Neutral Lipid Stain, is a kit designed to detect lipid droplets within cells using the highly sensitive and precise fluorescent probe LD540. This kit is suitable for use with fluorescence microscopes, flow cytometers, fluorescent plate readers, and other fluorescence detection systems.
Lipid droplets, composed of a hydrophobic core of neutral lipids such as triglycerides and cholesteryl esters, surrounded by a phospholipid monolayer, are widely present in animal, bacterial, yeast, plant, and insect cells. These droplets move along the cytoskeleton and interact with other organelles, playing crucial roles in lipid metabolism and storage, membrane transport, protein degradation, and signal transduction. Abnormal lipid storage is associated with various metabolic diseases such as obesity, fatty liver disease, cardiovascular diseases, diabetes, neutral lipid storage diseases, and Niemann-Pick C disease.
The fluorescent probe used in this kit is LD540 (4,4-Di-fluoro-2,3;5,6-bis-(tetra-methylene)-4-bora-3a,4a-di-aza-s-indacene). LD540 is a novel, highly sensitive lipophilic dye based on the BODIPY fluorescent group, achieving excellent co-localization with green fluorescent BODIPY 493/503 (Figure 2), and is suitable for precise lipid droplet labeling and imaging. LD540 features good photostability and suitable fluorescence spectra, making it compatible with common fluorescent dyes such as GFP, DAPI, and Alexa Fluor 647, supporting multicolor imaging needs. Additionally, LD540 is suitable for both fixed and live cells and can label ultra-small lipid droplets.
The molecular formula of LD540 is C18H21BF2N2, with a molecular weight of 314.18 and CAS number 151486-58-7. In ethanol, its maximum excitation wavelength is approximately 537nm, and its maximum emission wavelength is approximately 544nm.

Storage
- Store at -20ºC for at least one year.
- Store LD540 (1000X) and Hoechst 33342 (1000X) protected from light.
Precautions
- For the first use, aliquot LD540 (1000X) appropriately to avoid repeated freeze-thaw cycles.
- Optimize the working concentration of LD540, cell amount, incubation temperature, and time based on the cells used. For example, adjust the concentration of LD540 to 0.5X-2X for live cell staining and 0.1X-0.5X for fixed cell staining.
- LD540 is prone to quenching under laser exposure. Therefore, ensure to use the minimum dye concentration necessary to maintain fluorescence intensity, reduce the excitation light intensity of the fluorescence microscope, and shorten the exposure time.
- Fluorescent dyes are prone to quenching. Avoid light exposure to minimize fluorescence quenching.
