Description
Inclusion bodies are insoluble structures formed when recombinant proteins are overexpressed and do not have sufficient time to fold properly, leading to specific misfolding and aggregation of intermediate states. Although the amino acid sequence of these gene expression products is correct, their three-dimensional structure is incorrect, and therefore, they generally lack biological activity. Using this product to extract inclusion body proteins can increase the solubility of the inclusion body proteins and improve the extraction yield. Denaturants can be removed by dialysis.

Storage
Store at room temperature. This reagent is valid for six months from the date of purchase.
Notes
This reagent contains denaturants. Please wear a lab coat and disposable gloves while handling it.
Protocol
- Purify the inclusion bodies after lysing the bacterial culture using bacterial lysis solution or other methods.
- Add 8 ml of Inclusion Body Solubilization Reagent to 1 g of wet inclusion bodies (estimate the wet weight of inclusion bodies roughly).
- Mix thoroughly after adding the buffer and incubate with shaking for 30 minutes.
- Centrifuge to remove cell debris.
- Collect the supernatant for subsequent steps and testing.
