Human Cot‑1 DNA Pro is derived from genomic DNA extracted from human placenta. It consists of highly repetitive sequences approximately 50–300 bp in length. Description

Human Cot‑1 DNA Pro is derived from genomic DNA extracted from human placenta. It consists of highly repetitive sequences approximately 50–300 bp in length. These repetitive elements effectively block repetitive regions present in probes or target nucleic acids, thereby preventing nonspecific hybridization signals during nucleic acid hybridization. By masking repetitive sequences, Human Cot‑1 DNA Pro enhances hybridization specificity and improves the efficiency of target sequence capture.

Experimental results demonstrate that Human Cot‑1 DNA Pro significantly reduces false‑positive signals caused by repetitive sequences in chromosome in situ hybridization, lowers background noise, and improves the accuracy and reliability of hybridization assays. Most importantly, Human Cot‑1 DNA Pro features shorter and more uniform fragment sizes compared with conventional Human Cot‑1 DNA, resulting in superior suppression of nonspecific DNA binding.

 

Application

Human Cot‑1 DNA Pro is widely used in:

  • Northern blotting
  • Southern blotting
  • In situ hybridization, including tissue ISH, somatic cell ISH, and FISH
  • Liquid‑phase hybridization capture

Additionally, Human Cot‑1 DNA Pro can be used as a library screening probe for identifying clones in somatic cell hybrid libraries or flow‑sorted chromosome libraries derived from somatic cell hybrids.

 

Superior Blocking Performance

  • Utilizes a proprietary fragmentation process to ensure that ≥50% of DNA fragments fall within the 50–300 bp range.
  • Achieves a blocking efficiency of over 95%, significantly outperforming comparable products.
  • Effectively blocks common repetitive sequence families such as Alu and LINE.
