Description
Streptavidin (HRP-labeled Streptavidin), also known as HRP-Streptavidin, Streptavidin-HRP, or biotinylated peroxidase-labeled streptavidin, can be used for the detection of biotin-labeled antibodies, nucleic acids, proteins, or other biotinylated molecules. Specific applications include Western blotting, ELISA, immunohistochemistry or immunocytochemistry, in situ hybridization, Southern blotting, Northern blotting, EMSA, and more.
The molecular weight of Streptavidin is 52 kDa, and it can bind highly specifically to biotin. The affinity constant (Kd) between Streptavidin and biotin is in the range of 10^-15 M. Streptavidin is a tetrameric protein that can simultaneously bind to four biotin molecules. It shares a high degree of similarity in spatial structure and biotin affinity with Avidin obtained from chicken egg whites. Unlike Avidin, Streptavidin is a non-glycosylated protein and is essentially uncharged. Avidin has a pI of ~10.5, making it alkaline under neutral pH conditions. Due to its lack of charge under neutral conditions, Streptavidin exhibits much lower nonspecific binding compared to Avidin, reducing nonspecific background in detection assays. Therefore, Streptavidin is commonly used in biotin detection instead of Avidin.
Horseradish peroxidase (HRP), in the context of this HRP-labeled Streptavidin, catalyzes chemiluminescence reactions (such as ECL, BeyoECL, etc.) in Western blotting, EMSA, Southern or Northern blotting, producing a chemical glow. It can catalyze TMB to generate a blue color in ELISA and catalyze DAB to produce a brown precipitate in immunohistochemistry, immunocytochemistry, or Western blotting.
This HRP-labeled Streptavidin is produced by cross-linking high-purity Streptavidin with ultra-pure horseradish peroxidase, ensuring minimal background and maximum sensitivity in detection assays.
