Description

HEPES Solution (1M, pH 7.3, Cell Culture Tested) is prepared using high-purity HEPES (free acid) and high-quality NaOH, diluted with cell culture-grade ultrapure water, and sterilized by filtration. These solutions have a concentration of 1M and a pH of 7.3. They are commonly used as buffering agents in cell culture to maintain a stable pH range over an extended period.

HEPES (free acid), also known as 4-(2-Hydroxyethyl)piperazine-1-ethanesulfonic acid, has a molecular formula of C8H18N2O4S and a molecular weight of 238.30. Its CAS number is 7365-45-9. HEPES is a commonly used zwitterionic buffer reagent with an effective buffering range of pH 6.8-8.2 and a pKa value of 7.5 at 25°C.

HEPES Solution (1M, pH 7.3) is widely used in cell culture at a working concentration of 10-25 mM. Compared to bicarbonate buffer systems with lower pKa values, HEPES has a pKa of 7.5 and exhibits good buffering capacity within the pH range of 7.2-7.6. It is suitable for cell culture that requires precise pH conditions and provides excellent buffering capacity when cells are cultured in open systems or when observing cells for an extended period outside a CO2 incubator.

HEPES is often the preferred choice for buffering cell culture systems because it is impermeable to cell membranes, minimally affects various biochemical reactions, and exhibits stability in chemical and enzymatic environments. Additionally, HEPES has very low absorbance of visible and UV light.

HEPES is non-toxic to cells at concentrations of 10-25 mM. However, concentrations above 40 mM may become toxic to certain cells and affect cell growth.

 

Additional Notes

  • This product has a HEPES concentration of 1M, and the recommended working concentration of HEPES for cell culture is generally 10-25mM. The most commonly used concentration is 10mM, which can be used at a 100X dilution.
  • This product is sterile and has a pH of approximately 7.3±0.1 (25°C).
  • This product is intended for scientific research purposes by professionals only and is not to be used for clinical diagnosis or treatment. It is not intended for use in food or drugs and should not be stored in a regular residential setting.
  • For your safety and health, please wear laboratory attire and disposable gloves when handling.

 

Storage

Store at 4°C, effective for one year. Store at room temperature, effective for at least two weeks.
