FITC‑labeled Dextran Sulfate Sodium, abbreviated FITC‑DSS, also known as FITC‑Dextran Sulfate Sodium Salt, FITC‑labeled Dextran Sulfate (sodium salt), Fluorescein isothiocyanate–Dextran Sulfate (sodium salt), or Fluorescein‑labeled Dextran Sulfate (sodium salt), is a widely used fluorescent tracer compound commonly applied in the establishment of colitis animal models.
Dextran Sulfate Sodium (DSS) is a sulfated derivative of dextran. It is typically produced as a white powder by reacting dextran of defined molecular weight with sulfating agents such as chlorosulfonic acid, followed by purification. The DSS‑induced model is considered the gold standard for generating inflammatory bowel disease (IBD) models, and is widely used to induce colitis in mice, rats, zebrafish, and Drosophila. Its mechanism involves disrupting gut microbiota homeostasis, impairing macrophage function, and exerting cytotoxic effects on colonic epithelial cells. During use, DSS is often conjugated with fluorescein isothiocyanate (FITC) to visualize its distribution and potential sites of action. For example, FITC‑labeled DSS enables clear microscopic observation of tissue permeability changes in the colonic epithelium shortly after DSS exposure. FITC‑labeled DSS of different molecular weights also helps researchers rapidly assess cell membrane pore size. In addition, when analyzing mucus barrier alterations in IBD patients, FITC‑labeled DSS can be used to evaluate how DSS penetrates the mucus plume.

The labeling efficiency corresponds to approximately 0.1–2 FITC molecules per 100 glucose units, equivalent to 0.001–0.02 moles of FITC per mole of glucose.
This FITC‑labeled Dextran Sulfate Sodium series appears yellow to orange and exhibits high solubility in water, saline solutions, and many organic solvents such as DMSO, formamide, ethylene glycol, glycerol, and hexamethylphosphoramide. It is insoluble in lower aliphatic alcohols, acetone, chloroform, and dimethylformamide.
