Description
FITC-labeled Dextran, also known as FITC-Dextran, Fluorescein-labeled Dextran, Fluorescein-Dextran, FITC-labeled Dextran, Fluorescein isothiocyanate-Dextran, or Fluorescein-labeled Dextran, is a commonly used fluorescent tracer compound. It is frequently used in studies involving cell membrane permeability, blood vessels, intestines, blood-brain barrier, and other areas related to integrity and permeability, as well as in research related to cellular endocytosis.
Dextran, also known as (3’,6’-dihydroxy-3-Oxospiro(isobenzofuran-1-(3H), 9’-[9Hxanthen]-5 (or 6)-ylcarbamothiate), is a polymer of glucose, primarily composed of linear long molecular chains of D-pyranose glucose linked by α-1,6 glycosidic bonds, and branched molecular chains linked by α-1,2, α-1,3, and α-1,4 glycosidic bonds (Figure 1), with branches accounting for about 5%. Dextran is a hydrophilic polysaccharide with a wide molecular weight range (1kDa-40,000kDa), good water solubility, low toxicity, and low immunogenicity. Dextran is biologically inert and linked by the uncommon α-1,6 glycosidic bond, making it resistant to cleavage by most endogenous cellular glycosidases. Therefore, Dextran conjugates can serve as ideal long-term tracers for live cells.
Common dextran conjugates include fluorescently labeled dextran (such as FITC-Dextran) and biotinylated dextran. Their applications include, but are not limited to:
- Studying the size and permeability of cell membrane channels using fluorescently labeled dextran of different molecular weights.
- Research on plant cell wall permeability.
- Investigating ischemia-reperfusion injury, vascular permeability, blood-brain barrier integrity, and the function of organs such as the kidneys and intestines through perfusion experiments.
- Serving as effective anterograde and retrograde tracers in combination with neuroanatomical techniques to study the interactions of synapses at various levels of dendritic tissue.
- Determining cell structures such as gap junctions and cytoplasmic bridges by detecting the diffusion of fluorescently labeled dextran between cells.
- Research on cellular endocytosis.
- Dextran conjugates have been studied for the treatment of various diseases (e.g., tumors, liver diseases). Conjugation with dextran can enhance drug efficacy, improve cytotoxicity, and play a crucial role in controlled drug release and targeting.
- Some fluorescently labeled dextrans (e.g., FITC-labeled dextran) are pH-sensitive and can be used to monitor acidification.
- Studying the hydrodynamic properties of the cytoplasmic matrix.
- Research on wound healing processes, neuronal and vascular development during embryogenesis.
Our FITC-labeled dextran products are provided in lyophilized powder form, purified through precipitation, dialysis, and gel filtration, and tested by thin-layer chromatography (TLC) to ensure no free dye is present. These products are of high purity, excellent quality, and very stable, with a wide range of molecular weights available. Users can choose based on experimental purposes and required molecular weights.
Our FITC-labeled dextran products use fluorescein isothiocyanate (FITC) to label dextran of different molecular weights through a stable thiocarbamoyl linkage. The labeling process ensures that the dextran is not degraded and that the degree of FITC labeling on the polysaccharide chain is moderate, ensuring appropriate fluorescence intensity while minimizing potential impacts on the properties of dextran and avoiding interactions with intracellular components.
FITC is a commonly used green fluorescent probe that emits a green fluorescence signal upon excitation. Therefore, FITC-labeled Dextran can be detected using fluorescence microscopy, a fluorescence plate reader, or flow cytometry. The absorption (excitation) and emission peaks of FITC are shown in the table below.

The labeling efficiency of our FITC-labeled Dextran products is approximately 0.003-0.02 moles of FITC per mole of glucose, equivalent to 0.3-2 FITC molecules per 100 glucose molecules.
Our FITC-labeled Dextran products are yellow or orange in color and have high solubility in water, saline solutions, and many organic solvents (such as DMSO, formamide, ethylene glycol, glycerol, and hexamethylphosphoramide), but are insoluble in lower alcohols, acetone, chloroform, and dimethylformamide.
Storage
Store at -20ºC, protected from light. Valid for at least one year.
