Description
Exonuclease VIII, Truncated, also known as truncated Exonuclease VIII, originates from Escherichia coli. Exonuclease VIII, truncated, has 5’ to 3’ DNA exonuclease activity, specifically cleaving linear double-stranded DNA. It exhibits similar enzymatic activity on 5’ ends that are blunt or sticky, and on 5’ ends with either phosphate or hydroxyl groups. However, Exonuclease VIII, truncated, cannot cleave circular DNA. Experimental results show that Exonuclease VIII, truncated, can digest single-stranded DNA with 5’ ends that are either phosphorylated or hydroxylated from the 5’ to 3’ direction, but its activity on single-stranded DNA with 5’ phosphorylated ends is significantly higher than on those with 5’ hydroxylated ends.
Application
Digests linear double-stranded DNA while maintaining circular double-stranded DNA.

Unit Definition
One unit is defined as the amount of enzyme required to produce 1 nmol of acid-soluble deoxyribonucleotide from double-stranded DNA in a total reaction volume of 50 µl in 30 minutes at 37ºC in 1X Exonuclease VIII, truncated Buffer with 0.15 mM sonicated duplex [3H] DNA.
Purity
Free from other types of exonucleases, endonucleases, RNases, and phosphatases, except for Exonuclease VIII, truncated.
Inactivation or Inhibition
Heating at 70ºC for 15 minutes will inactivate Exonuclease VIII, truncated.
Storage
Store at -20ºC, effective for at least two years.
