Description
Enhanced Exosome Isolation Kit from Cell Culture Media (Precipitation Method), also known as Exosome Purification Kit, Exosome Extraction Kit, or Total Exosome Isolation Kit, is an efficient and convenient kit for extracting exosomes or other extracellular vesicles from cell culture supernatants. It allows the isolation of a large number of intact, high-purity exosomes with simple low-speed centrifugation. The exosomes extracted using this kit can be used for protein analysis, nucleic acid analysis, Western blot, PCR, RNA extraction, high-throughput sequencing, exosome particle size and concentration analysis (Nanoparticle Tracking Analysis, NTA), electron microscopy analysis, and cell co-culture experiments.
The yield of this kit is significantly improved. Compared to the standard Exosome Isolation Kit from Cell Culture Media (Precipitation Method), this enhanced kit has further optimized the formulation, greatly improving the efficiency and stability of exosome extraction. In most cases, the actual yield can be increased by at least three times.
Exosomes are membrane-bound extracellular vesicles (EVs) with a diameter of approximately 40-160 nm and a lipid bilayer structure. They naturally exist in blood, urine, cerebrospinal fluid, and the supernatant of cultured cells. Almost all types of cells can produce and release exosomes. Endocytosis of the cell membrane sequentially forms early sorting endosomes (ESE), late sorting endosomes (LSE), and multivesicular bodies (MVB), which contain intraluminal vesicles (ILVs). MVBs fuse with the cell membrane to form exosomes, which carry various components from their parent cells (including nucleic acids, proteins, lipids, carbohydrates, and metabolites) and release them into the extracellular matrix. Exosomes can be recognized and fused by nearby or distant cells, serving as important mediators of intercellular regulation. They are involved in the pathogenesis of various diseases such as cancer, neurodegenerative diseases, and inflammatory diseases, affecting multiple cellular functions.
This product uses an improved polymer precipitation method, requiring low equipment demands, easy operation, short extraction time, low sample starting amount, high extraction efficiency, and relatively intact exosome morphology.
Compared to the standard Exosome Isolation Kit from Cell Culture Media (Precipitation Method), this enhanced kit further optimizes the formulation. During the exosome separation and extraction process, it reduces the adsorption effect of hydrophilic or hydrophobic groups and surface charges on consumable materials such as centrifuge tubes and pipette tips. This minimizes significant losses of exosomes during extraction, thereby improving exosome extraction efficiency and stability.
This product is free of RNase, DNase, and protease. The exosomes extracted using this product can be used for various RNA, DNA, and protein applications.
For most cell culture supernatants, only 10 ml of cell culture supernatant is needed to obtain exosomes through a 2-hour precipitation, making the process relatively time-saving and labor-efficient. For cells that secrete fewer exosomes, the exosome extraction rate can be improved by increasing the volume of cell culture supernatant or extending the precipitation time.
