Double-stranded DNA (dsDNA) Fragmentase is a time-dependent enzyme that can cleave dsDNA into fragments of 50-1,000 bp depending on the reaction time. dsDNA fragmentase consists of two enzymes: one enzyme randomly nicks the dsDNA, and the other enzyme recognizes these nicks and cuts the strands, generating dsDNA fragments. The resulting DNA fragments have very short overhangs, with 5' phosphorylated ends and 3' hydroxyl ends. Library preparation and sequencing validation have shown that EVNext dsDNA fragmentase has random cleavage characteristics. Comparing libraries prepared with this enzyme to those prepared by mechanical methods, sequencing results showed no bias, and there was no difference in sequencing coverage.
