Description
DNA Size Selection Magnetic Beads (DSM Beads) are based on the Solid Phase Reverse Immobilization (SPRI) technology using novel nucleic acid purification media coated magnetic beads. They, along with an optimized buffer system, allow for the isolation and purification of specific length range DNA or RNA fragments from a suspension of magnetic beads at a specific ratio. This product is suitable for recovering, separating, or purifying DNA or RNA fragments within a specific length range greater than 100 base pairs (bp). It is particularly useful for length selection of DNA or RNA during high-throughput sequencing library construction.
By adjusting the amount of this product and performing single-sided or double-sided (one or two rounds) separation and purification, different length range DNA can be achieved.
Nucleic acid purification and removal of impurities are crucial for genome-related experiments, including sequencing, qPCR/ddPCR/PCR, microarrays, and other enzyme-catalyzed reactions. This product finds wide applications in areas such as genetic sequencing and molecular diagnostics, especially in the length selection and purification of DNA or RNA during the process of next-generation sequencing (NGS) library construction. It is compatible with conventional library preparation kits and exhibits consistent usage, fragment recovery rate, and size distribution with the widely used Agencourt AMPure XP Beads (a product by Beckman Coulter). DNA size selection magnetic beads are also known as DNA size selection purification beads, DNA fragment selection beads, DNA selection beads, DNA screening beads, DNA clean beads, NGS cleanup and size select, DNA selection beads, DNA select isolation kit, sample purification beads, and so on.
The working principle of this product is as follows: the product consists of a salt solution containing magnetic beads and substances like polyethylene glycol (PEG). In the presence of a certain concentration of salt and PEG, the DNA molecule undergoes conformational changes, exposing a large number of phosphate groups that bind to carboxyl-coated magnetic beads. Subsequently, through magnetic separation, after removal of PEG and salt, DNA can be eluted from the magnetic beads, obtaining purified DNA that has undergone separation. By controlling the concentration of PEG and salt in the buffer solution during the experimental process, different sizes of DNA fragments can be bound to the magnetic beads and purified.
Features
- This product exhibits strong specificity and high DNA binding capacity. It enables fast DNA binding and high binding capacity, allowing for rapid separation and purification of DNA fragments of different sizes. Additionally, the magnetic beads have small particle size, reducing non-specific adsorption and enabling quick and effective removal of unincorporated dNTPs, primers, primer dimers, salts, and other contaminants.
- The product demonstrates broad compatibility. It is suitable not only for manual operations with small sample sizes but also for high-throughput automated systems.
- The length selection range provided by this product is convenient and efficient. By adjusting the amount of magnetic beads, the range of DNA length selection can be modified to accommodate different experimental needs. Precautions
- Although this product is similar to other similar products from different companies in terms of usage instructions and fragment recovery rates, it is still recommended to perform some testing and optimization.
- Avoid freezing and high-speed centrifugation when using this product.
- To ensure the recovery rate of DNA, it is recommended to remove the magnetic beads from the refrigerator at 4°C before use or take the required amount and allow them to equilibrate to room temperature.
- When aliquoting or using the magnetic beads, it is advisable to vortex or invert them adequately to ensure thorough mixing.
- It is recommended to prepare an 80% (v/v) ethanol solution just before use, as prolonged storage may lead to ethanol evaporation and affect the recovery efficiency.
- The initial volume of DNA samples should be greater than 100 μl. If the volume is insufficient, it can be supplemented with ultrapure water to reach 100 μl. Insufficient sample volume can result in uneven incubation with the magnetic beads, increased pipetting errors, and subsequently affect the accuracy of the size selection.
- Do not let the magnetic beads dry for an extended period, as excessive drying can irreversibly aggregate the beads and reduce elution efficiency.
- This product is intended for scientific research by professionals only and should not be used for clinical diagnosis or treatment, food or drug purposes, or stored in regular residential areas.

Storage
Store at 4°C, valid for one year.
