Concanavalin A Magnetic Beads, also referred to as ConA beads, Concanavalin A beads, concanavalin A-coated beads, concanavalin-coated beads, concanavalin A-conjugated beads, or concanavalin protein A beads, are formed by covalently coupling high-quality Concanavalin A (ConA) to superparamagnetic nanoparticles. These beads possess the capability to rapidly, efficiently, sensitively, and specifically bind to molecules with glycosylation modifications such as glycoproteins, glycolipids, and polysaccharides. They are primarily utilized for experiments involving the separation of glycosylated molecules, including glycoproteins, from cells, tissue lysates, or serum samples. Notably, they can be employed in Chromatin Unbiased Targeted CUT&RUN assays for precise targeting, cleavage, and release of DNA with the assistance of nucleases.
Concanavalin A, abbreviated as ConA or Con A, known as Concanavalin A in Chinese, also referred to as concanavalin A-associated protein, jack bean protein A, or jack bean agglutinin A, is a lectin isolated from Jack beans (Canavalia ensiformis). It is a mannose/glucose-specific agglutinin and is one of the most widely used lectins. Its CAS number is 11028-71-0, and the monomer has a molecular weight of approximately 25.5 kDa. It can bind to one Ca2+ and one Mn2+ ion and contains a sugar-binding site with high affinity for terminal α-D-mannose and α-D-glucose residues. Concanavalin A is a T-cell mitogen that can activate the immune system, recruit lymphocytes, and induce cytokine production. Apart from its mitogenic activity, Concanavalin A can also induce programmed cell death through specific molecular mechanisms. It has been reported that Concanavalin A can activate NFAT (Nuclear factor of activated T cells), a transcription factor family that plays a crucial role in the development and function of the immune system. Concanavalin A exists as a tetramer when the pH is between 5.8 and 7.0. It dissociates into a dimer under conditions of pH 6.5 or lower, and at pH above 7.0, it can form larger aggregates.
ConA magnetic beads find extensive applications in the fields of immunology, biochemistry, and medicine. They are used for cell separation, as well as the isolation of glycosylated molecules such as glycoproteins from cell lysates, tissue lysates, or serum, including in novel techniques like Cleavage Under Targets & Release Using Nuclease (CUT&RUN) for chromatin analysis. ConA magnetic beads play a critical role in these applications. The use of ConA magnetic beads for cell separation or fixation is convenient and minimizes cell loss during subsequent washing steps. Additionally, ConA magnetic beads can be employed for the collection and fixation of cell nuclei. The experimental procedure for purifying glycoproteins using ConA magnetic beads is depicted in Figure 1, and the process for cell or nucleus separation/fixation is similar.

- This product exhibits strong specificity. It possesses a highly specific recognition capability for carbohydrates, enabling selective binding to carbohydrates. It can separate molecules such as cells, cell nuclei, or glycoproteins with appropriate glycosylation modifications. It can be directly utilized in experiments like CUT&RUN, providing glycosylated proteins that can be analyzed and detected through Western blotting and mass spectrometry.
- This product offers a high binding capacity. Compared to similar products, it demonstrates a high binding capacity for glycoproteins. Additionally, the small size of the magnetic beads minimizes nonspecific adsorption. Each milliliter of magnetic bead suspension contains ≥1 mg of ConA, with each ConA protein having four sugar-binding sites. Thus, each milliliter of magnetic bead suspension can bind ≥1 mg of glycoproteins, enabling efficient experiments such as cell or glycoprotein separation and purification.
- This product exhibits rapid glycoprotein binding with a short adsorption time, facilitating efficient carbohydrate binding. The nanoparticles used in this product (~200nm) possess an ultra-large surface area, which effectively reduces the time required for ConA-carbohydrate binding. Shortening the operation time prevents glycoprotein degradation during extended procedures, ensuring the activity and integrity of glycoproteins.
- This product is convenient to use. It is stored in a special protective solution without glycerol. It employs magnetic separation, eliminating the need for centrifugation. This product is suitable not only for detecting small sample quantities but also for automated high-throughput screening systems, ensuring high consistency across different operating methods.
