Description
Circular RNA Synthesis Kit, also known as the Circular RNA Preparation Kit, is a convenient and efficient enzymatic method for converting single-stranded RNA (ssRNA) into circular RNA (circRNA). The synthesized circRNA can be used for studying the biological functions of circRNA, including transfection into cells. Recent studies have shown that circRNA can also be used as mRNA vaccines, including vaccines for viruses such as the novel coronavirus (2019-nCoV or SARS-CoV-2).
Circular RNA is a type of RNA molecule that forms a circular structure through typical 5'-3' phosphodiester bonds. This structural feature makes it resistant to degradation by RNA exonucleases, resulting in greater stability and resistance to degradation. CircRNA is a naturally occurring single-stranded circular RNA commonly found in organisms and is generated through processes such as intron reverse splicing (trans-splicing).
The study of circRNA can be done through plasmid or viral expression methods, utilizing processes such as reverse splicing to generate circRNA. However, the use of plasmids or viruses for circRNA generation may involve auxiliary processes such as reverse splicing, which can potentially interfere with research results. Direct transfection of synthesized circRNA allows for a more direct investigation of the biological functions of circRNA.
The principle of the kit for the circularization of single-stranded RNA involves the use of an RNA cyclase enzyme to cyclize the 5'-phosphorylated and 3'-hydroxylated ends of the RNA molecule.
Feature
High circularization efficiency and easy to use: The circularization efficiency typically reaches 95% or higher, and the procedure can be completed with a single incubation step. With this kit, more than 95% of 5'-phosphorylated and 3'-hydroxylated single-stranded RNA (ssRNA) can be converted into circular RNA (circRNA). As a result, the circularization products may not require gel purification. After heat inactivation of the cyclase enzyme, the products can be used for subsequent experiments by simply precipitating with ethanol.
Storage
The minimum shelf life is 1 year at -20°C.
Note
- The 3' end of the substrate single-stranded RNA must be hydroxylated, while the 5' end can be phosphorylated or adenylated.
- The circularization reaction system provided by this kit can be scaled up proportionally without significantly affecting the yield of circular RNA products.
