Description

The ​Cell-Free Protein Synthesis Kit​ is an in vitro protein production system based on E. coli cell lysate. This product utilizes active components in the lysate—such as ribosomes, translation factors, and enzymes—while supplementing with energy sources, nucleotides, amino acids, and inorganic salts to reconstruct a transcription-translation system outside of living cells. Using DNA or RNA as a template, it enables efficient protein expression.

Cell-free protein synthesis offers a rapid, flexible, and scalable approach to producing proteins without relying on living bacterial systems. Compared to traditional recombinant expression methods, it provides several key advantages:

  1. ​Fast protein expression: Target proteins can be expressed within just 1–2 hours, reaching maximum yield in 8–24 hours.
  2. ​High protein yield: Protein concentrations can reach up to ​3 mg/mL​ or higher.
  3. ​Simple and flexible workflow: Just add your DNA or RNA template to the reaction mixture. The system is compatible with standard formats such as ​96-well plates or centrifuge tubes.

​Note: This product (under the current catalog number) is ​not suitable for soluble expression of proteins containing disulfide bonds. If you require soluble expression of such proteins, please inquire about alternative catalog numbers.

 

Principle

The Escherichia coli cell-free protein expression system is currently the most widely used among cell-free protein expression systems. Due to its well-understood genetic background, high expression levels of target genes, short cultivation cycles, high efficiency, low cost, strong resistance to contamination, ease of scale-up, and applicability to the expression of proteins from various species, it is extensively applied in cutting-edge research fields such as protein-protein interactions, high-throughput screening, and gene circuits. Utilizing an efficient transcription and translation coupling reaction, this system produces active recombinant proteins in vitro. The highly efficient and rapid cell-free synthesis system eliminates time-consuming steps in traditional intracellular protein production processes, including transformation, cell cultivation, and expression optimization. Within a few hours, this E. coli cell-free expression system can generate high yields of high-quality proteins. Whether using circular plasmid DNA for protein synthesis, rapid expression screening, or downstream structural or functional analysis, this system significantly simplifies the protein expression process.

Store at –20°C for up to 12 months.

After reconstitution of the lyophilized powder:
