Description
The Cell‑Free Protein Synthesis Kit is an E. coli lysate–based in vitro protein expression system. This version is designed for the site‑specific incorporation of the non‑canonical amino acid Nε‑crotonyl‑L‑lysine (Kcr) by utilizing the amber stop codon (TAG) within the DNA sequence. The kit includes three additional components—Kcr, tRNA, and aminoacyl‑tRNA synthetase—all of which can be adjusted and optimized according to the user’s experimental requirements.
Note: This product is not suitable for soluble expression of proteins containing disulfide bonds, as the reaction system does not provide an oxidative environment that supports correct disulfide bond formation. For soluble expression of disulfide‑bonded proteins, please refer to other catalog numbers.
Principle
The Escherichia coli cell-free protein expression system is currently the most widely used among cell-free protein expression systems. Due to its well-understood genetic background, high expression levels of target genes, short cultivation cycles, high efficiency, low cost, strong resistance to contamination, ease of scale-up, and applicability to the expression of proteins from various species, it is extensively applied in cutting-edge research fields such as protein-protein interactions, high-throughput screening, and gene circuits. Utilizing an efficient transcription and translation coupling reaction, this system produces active recombinant proteins in vitro. The highly efficient and rapid cell-free synthesis system eliminates time-consuming steps in traditional intracellular protein production processes, including transformation, cell cultivation, and expression optimization. Within a few hours, this E. coli cell-free expression system can generate high yields of high-quality proteins. Whether using circular plasmid DNA for protein synthesis, rapid expression screening, or downstream structural or functional analysis, this system significantly simplifies the protein expression process.

Storage and Handling
- Store at –80 °C with a 12‑month shelf life.
- Ship on dry ice. After opening, keep all cell‑free protein expression components at –80 °C.
- To avoid activity loss caused by repeated freeze–thaw cycles, aliquot Solution A and Solution B according to your reaction scale. Snap‑freeze the aliquots in liquid nitrogen, then transfer them to –80 °C for long‑term storage.
