Description
Carrier RNA (Poly A), also known as carrier RNA (Poly A) or carrier polyadenylic acid, is a mixture composed of polyadenylic acid (Poly A) strands ranging from approximately 100 to 10,000 nucleotides in length. It is also referred to as a nucleic acid coprecipitant or potassium polyadenylate. This substance is primarily used to enhance the recovery or extraction efficiency of DNA and RNA from trace samples, such as viral samples, short nucleic acids (less than 200 bp), or small RNAs.
Carrier RNA (Poly A) is suitable for most viral DNA/RNA extraction kits. On one hand, it can improve the binding and elution efficiency of DNA/RNA during the purification process for trace samples. On the other hand, it reduces the likelihood of degradation by residual nucleases. The effect of carrier RNA is more pronounced when the sample nucleic acid content is low. Additionally, common centrifuge tubes are made of polypropylene (PP), whose walls may have static electricity that can adsorb trace nucleic acids. By directly adding Carrier RNA (Poly A) to the lysis buffer during RNA extraction, the carrier RNA can be effectively adsorbed onto the purification column, removing the static electricity interference and improving extraction efficiency.
This Carrier RNA (Poly A) is synthesized in vitro through the polymerization of adenylic acid by polynucleotide phosphorylase (PNPase) and is not derived from microorganisms, viruses, or cells, thereby not interfering with subsequent qPCR or other DNA/RNA detection methods.
The CAS number for Carrier RNA (Poly A) is 26763-19-9, with a purity of approximately 99%. The strand length ranges from about 100 to 10,000 nucleotides, mainly between 100 and 2,000 nucleotides, and the molecular weight is approximately 35 to 3,500 kDa.
This Carrier RNA (Poly A) is provided in the form of a lyophilized powder.
Storage
Store at -20ºC, effective for two years.
Precautions
- Handle with care to avoid contamination. Use RNase-free pipette tips and Eppendorf tubes. It is recommended to wear a disposable mask while handling.
- For samples with small quantities or when extracting viral DNA/RNA, it is advisable to use Carrier RNA (Poly A). If a large amount of DNA/RNA is expected, you may choose whether to add Carrier RNA (Poly A) based on your needs.
- Due to the presence of Carrier RNA (Poly A), the concentration of viral DNA/RNA cannot be directly quantified using absorbance or conventional fluorescent staining methods. It is recommended to use qPCR or qRT-PCR for quantification.
