Description
Reticulocytes are transitional cells between late-stage erythrocytes and fully mature red blood cells. Due to the presence of alkaline RNA material in their cytoplasm, a live stain with Brilliant Cresyl Blue solution reveals a reticulated structure in the cytoplasm, appearing as light blue or deep blue under microscopic examination. The staining results obtained with this solution exhibit a bright and clear background, unaffected by prolonged staining. This staining solution is intended for research purposes only and is not to be used for clinical diagnosis or other applications.
Materials Needed
- Fresh whole blood or EDTA anticoagulated whole blood
- Hemocytometer
- Microscope
Operating Steps (for reference only)
- Mix Brilliant Cresyl Blue staining solution with whole blood at a 1:1 ratio, let it stand at room temperature for 10-15 minutes or longer.
- Prepare blood smears as usual.
- Observe under a microscope or count at least 1000 red blood cells, including reticulocytes.
Staining Results
Reticulocytes exhibit a reticulated structure with light blue or deep blue staining in the cytoplasm.
Calculation
Reticulocyte Percentage = (Count of reticulocytes in 1000 red blood cells / 1000) x 100
Reticulocyte Absolute Count (cells/L) = Reticulocyte Percentage x Red blood cell count / L
Notes
- Ensure sufficient staining time, and do not prepare smears immediately after mixing. When the room temperature is low, the staining time should be extended accordingly.
- Blood smears should be evenly thick, with no overlap of red blood cells to avoid affecting the staining effect.
- Fresh whole blood or EDTA-anticoagulated blood is required for blood cell smear staining.

Store at room temperature.
