Description
Broad-spectrum nuclease Benzonuclease is a non-specific endonuclease derived from Serratia marcescens, also known as "red bacteria", which has been modified using protein engineering techniques. It does not contain bacterial endotoxins expressed by prokaryotes. Functionally, it can cleave various forms of DNA and RNA, including single-stranded, double-stranded, linear, circular, or supercoiled DNA and RNA, by breaking phosphodiester bonds to produce 5’-phosphate nucleotides or 5’-phosphate oligonucleotides. Benzonuclease does not require specific nucleic acid base sequences, and can cut between any nucleotides in a chain, making it suitable for removing nucleic acids from biological products. In vaccine production, protein, and polysaccharide pharmaceutical industries, it is widely used to remove residual host nucleic acids, enabling nucleic acid contamination in vaccines and protein products to reach the picogram (pg) level. Benzonuclease can also be used as a preferred enzyme preparation for reducing viscosity in cell culture supernatants and cell lysates.

- Microbial Criteria: After solid enzyme is dissolved, if storage is required, it is recommended to be filtered and stored at -20°C. Liquid enzyme, after sterilization by 0.22μm filtration, shows no microbial contamination.
- Activity Assay and Definition of Activity Unit: One enzyme activity unit is defined as the amount of oligonucleotides generated in 30 minutes under reaction conditions at 37°C and pH 8.0, equivalent to a change of 1.0 in absorbance at 260 nm.
- Transportation, Storage, and Stability: Liquid enzyme is recommended for low-temperature transportation and storage. Solid enzyme can be transported and stored at room temperature for short periods. It is advisable to store sealed at temperatures below 4°C. The shelf life of liquid enzyme is one year. After opening, if left at 2-8°C for more than one week, it is recommended to filter sterilize to prevent microbial contamination. The storage life of solid enzyme is three years. After opening, use promptly and avoid moisture.
