Description
In normal cells, phosphatidylserine (PS) is only found on the inner leaflet of the cell membrane lipid bilayer. However, during the early stages of apoptosis, PS translocates from the inner to the outer leaflet of the cell membrane. Annexin V is a calcium-dependent phospholipid-binding protein with a molecular weight of 35-36 kD that has a high affinity for phosphatidylserine. As a result, it can bind to the exposed phosphatidylserine on the outer leaflet of the membrane in early apoptotic cells. Therefore, Annexin V is used as a sensitive marker for detecting early apoptosis. Annexin V is labeled with the fluorescent dye sFluor647, which acts as a fluorescent probe to detect apoptosis using fluorescence microscopy or flow cytometry.
This kit is suitable for detecting apoptosis in cultured cells and is not recommended for use with tissue samples.

Additional Notes
- Annexin V-sFluor647 components should be aliquoted as needed and stored at -20°C, avoiding repeated freeze-thaw cycles.
- Propidium iodide and Binding Buffer components can be stored at 4°C when not in use. Propidium iodide should be protected from light.
- Before using micro-reagents, please centrifuge to collect the liquid.
- Propidium iodide (PI) is toxic. Please wear gloves when handling it.
- This kit is suitable for detecting live cells. When using a flow cytometer, the cell count should not be lower than 1x105. It is not recommended for detecting tissue samples.
- Suspension-cultured cells are recommended. For adherent cells, use trypsin without EDTA for digestion. Improper digestion may cause false positives, and using a cell scraper may lead to cell clumping, affecting detection. Cells digested with trypsin can be stored in PBS containing 2% BSA to prevent further damage.
- Fixation of cells may cause fluorescence quenching; please do not fix the samples.
- Due to variations in cell types, apoptosis inducers, and detection instruments, fluorescence compensation for flow cytometry may vary. Therefore, it is recommended to use cells treated with apoptosis inducers as controls for fluorescence compensation adjustment in each experiment.
Storage
Store at 2-8°C for 12 months, protected from light.
