Description
3173 DNA Polymerase is a thermostable DNA polymerase derived from a virus. Through point mutation modification, its 3 '- 5' exonuclease activity is deleted (exo form) to obtain better amplification activity. This enzyme not only has DNA-dependent polymerase activity but also has RNA-dependent reverse transcription activity, so it can be used for single-tube RT-PCR amplification. The enzyme has strong chain replacement activity and can be used for the isothermal amplification of DNA or RNA. In the LAMP experiment using RNA as a template, a single enzyme system reaction can be realized. For RNA templates with complex secondary structures, a good amplification effect can be obtained by heating at 92°C for 3s, while Bst pol will lose amplification activity after heating at this temperature.
The half-life of 3173 DNA polymerase at 95°C is 20 min, so the initial pre-denaturation time of PCR is usually 1~2 min. This enzyme is matched with two kinds of Buffer: 10 × 3173 Isothermal Buffer and 5 × Best PCR Buffer。 10 × 3173 Isothermal Buffer is used for isothermal amplification, and its reaction temperature cannot exceed 70°C. 5 × Best PCR Buffer is used for routine PCR amplification, and the pre-denaturation time at 94°C cannot exceed 5min.
