PfAgo (Lyophilized)

PfAgo is an Argonaute protein derived from the hyperthermophilic archaeon Pyrococcus furiosus, exhibiting programmable endonuclease activity. Guided by a 5'-phosphorylated single-stranded DNA (gDNA), it specifically recognizes and cleaves complementary target DNA sequences. The guide DNA (gDNA) typically consists of a 16–18 nt, 5'-phosphorylated single-stranded oligonucleotide. The cleavage site is precisely located between the 10th and 11th nucleotides from the 5' end of the complementary guide sequence. This version is supplied in lyophilized form. It can be shipped at ambient temperature to reduce transportation costs.

sssLbCas12a Nuclease

sssLbCas12a (single-stranded DNA-specific LbCas12a) is a mutant engineered from wild-type LbCas12a endonuclease. Obtained via targeted engineering and screening, this mutant specifically recognizes target ssDNA while exhibiting extremely low activity for the recognition and cleavage of target dsDNA. Similar to the mechanism of action of wild-type LbCas12a, sssLbCas12a is crRNA-guided to recognize and cleave target ssDNA in a PAM-independent manner. In addition, recognition and binding to target ssDNA can activate the trans-cleavage activity of sssLbCas12a, degrading ssDNA probes present in the reaction system. Therefore, sssLbCas12a can be applied for rapid detection of target nucleic acids.

cis-Only FnCas12a Nuclease

cis-Only FnCas12a is a mutant engineered from wild-type FnCas12a endonuclease. Through targeted engineering and screening, this mutant retains only cis-cleavage activity against target DNA sequences; no significant trans-cleavage activity can be detected for this enzyme, regardless of whether the target is dsDNA or ssDNA. Therefore, this enzyme can be used in applications sensitive to Cas12a trans-cleavage activity.

enLbCas12a Nuclease

enLbCas12a (enhanced LbCas12a) is a high-activity LbCas12a mutant engineered from wild-type LbCas12a endonuclease. Sharing the same mechanism of action as wild-type LbCas12a, enLbCas12a is guided by crRNA to specifically recognize double-stranded DNA bearing the PAM sequence (5’-TTN-3’), resulting in double-stranded DNA breaks with sticky ends. Meanwhile, recognition and cleavage of single-stranded DNA targets by enLbCas12a are PAM-independent. Once enLbCas12a assembles into a ternary complex with crRNA and target DNA, it triggers cleavage of non-specific ssDNA sequences, degrading ssDNA present in the reaction system. GeCas12a2 Nuclease

GeCas12a2 nuclease was screened from environmental metagenomes rather than from a single isolated and purified known axenic strain. Its amino acid sequence shares 89% identity with SuCas12a2. Cas12a2 is a crRNA-guided endonuclease that targets RNA sequences, and the direct repeat (DR) sequence of its crRNA is largely consistent with that of Cas12a. Under the guidance of crRNA, GeCas12a2 assembles into a ternary complex together with crRNA and target ssRNA, enabling specific cleavage of the target ssRNA. Meanwhile, GeCas12a2 is activated to exert trans-cleavage activity toward unpaired ssDNA, ssRNA and dsDNA, indiscriminately degrading any ssDNA, ssRNA and dsDNA sequences present in the system. Unlike other Cas nucleases, which only perform trans-cleavage on ssDNA (e.g., Cas12a, Cas12b) or ssRNA (e.g., Cas13a) upon target activation, GeCas12a2 exhibits substantially stronger non-specific cleavage activity.

FnCas12a Nuclease (Lyophilized)

FnCas12a (also known as Cpf1) nuclease is derived from the strain Francisella tularensis. FnCas12a is a crRNA-guided DNA endonuclease. In the presence of a PAM (TTN) sequence on the target double-stranded DNA, it specifically cleaves the target double-stranded DNA, inducing double-strand breaks with sticky ends. In contrast, the specific cleavage of single-stranded DNA targets by FnCas12a is PAM-independent. Once FnCas12a assembles into a ternary complex with crRNA and target DNA, it triggers collateral cleavage of non-specific single-stranded DNA (ssDNA), degrading any ssDNA sequences present in the reaction system. For this reason, FnCas12a can be applied for rapid detection of target nucleic acids.
