Recombinant Human VEGF165
Vascular Endothelial Growth Factor is a sub-family of growth factors produced by cells, which stimulates vasculogenesis and angiogenesis. VEGF's normal function is to create new blood vessels during embryonic development, new blood vessels after injury, muscle following exercise, and new vessels (collateral circulation) to bypass blocked vessels. Humans express alternately spliced isoforms of 121, 145, 165, 183, 189, and 206 amino acids (a.a.) in length. VEGF production can be induced in cells that are not receiving enough oxygen. VEGF165 appears to be the most abundant and potent isoform, followed by VEGF121 and VEGF189. Recombinant human VEGF165 contains 166 amino acids residues and it is a disulfide-linked homodimer. In addition, it shares 88%a.a. with corresponding regions of mouse and rat, 96% with porcine, 95% with canine, and 93% with feline, equine and bovine VEGF, respectively.

This gene encodes a member of the epidermal growth factor superfamily. The encoded preproprotein is proteolytically processed to generate the 53-amino acid epidermal growth factor peptide. This protein acts a potent mitogenic factor that plays an important role in the growth, proliferation and differentiation of numerous cell types. Recombinant Human EGF
Epidermal Growth Factor (EGF) was originally discovered in crude preparations of nerve growth factor prepared from mouse submaxillary glands as an activity that induced early eyelid opening, incisor eruption, hair growth inhibition, and stunting of growth when injected into newborn mice. Human EGF was isolated from urine based on its inhibitory effect on gastric secretion and named urogastrone, accordingly. EGF is prototypic of a family of growth factors that are derived from membrane-anchored precursors. All members of this family are characterized by the presence of at least one EGF structural unit (defined by the presence of a conserved 6 cysteine motif that forms three disulfide bonds) in their extracellular domain. EGF is initially synthesized as a 130kDa precursor transmembrane protein containing 9 EGF units. The mature soluble EGF sequence corresponds to the EGF unit located proximal to the transmembrane domain. The superfamily of insulin-like growth factor (IGF) binding proteins include the six high-affinity IGF binding proteins (IGFBP) and at least four additional low-affinity binding proteins referred to as IGFBP related proteins (IGFBP-rP). Insulin-like Growth Factor-Binding Protein 3 (IGF-BP3) belongs to the IGFBP family, which are all cysteinerich proteins with conserved cysteine and have an IGFBP domain and a thyroglobulin type-I domain. Recombinant Human FGF-12
Fibroblast growth factor-12 is a member of the FGF superfamily of molecules contains at least 22 members. Human FGF-12 is synthesized as a 243 aa protein. It lacks a typical signal sequence and is considered to be a cytoplasmic protein. It does, however, possess an N-terminal bipartite nuclear localization signal (NLS) at aa 11 - 18 and 28 - 38. The 243 aa protein has at least one alternate splice form that is 181 aa in length. This is termed FGF-12B. Alternate splicing deletes the N-terminal 66 aa in FGF-12 and replaces them with four aa in FGF-12B. This substitution removes the NLS from the short form. Studies on the short form (12B) show that it cannot bind any of the common FGF receptors. This is consistent with its cytoplasmic localization. It can, however, bind to IB2 (islet brain-2), a cellular kinase scaffold protein, and voltage-gated sodium channels, suggesting FGF-12B plays an important role in intracellular signaling and ion exchange. Mouse and human FGF-12B differ by only one amino acid.
