Description
UniProof HiFi DNA Polymerase is a versatile, all-in-one enzyme developed through our proprietary molecular enzyme evolution platform (in silico Design & in vitro Evolution Workflow). It combines the strengths of Q5 and Q5U polymerases, delivering enhanced performance across diverse applications.
Derived from a high-fidelity DNA polymerase backbone, UniProof incorporates an optimized extension domain for robust long-fragment amplification and high yield. Through intensive screening of the exonuclease domain, it achieves exceptional fidelity—comparable to Q5—with 280× the fidelity of Taq polymerase. Engineering at the substrate-binding center confers tolerance to dUTP and enables efficient amplification of uracil-containing templates.
UniProof readily amplifies up to 8 kb of genomic DNA or 20 kb of λDNA, boasting an extension rate exceeding 2 kb/min. The enzyme exhibits strong 5’→3’ polymerase activity and robust 3’→5’ exonuclease (proofreading) activity, producing blunt-end products.
Unlike many B-family high-fidelity polymerases that stall at uracil residues in the template strand, UniProof lacks a uracil-binding domain, enabling it to read and amplify uracil-containing DNA. This unique “U-tolerant” feature supports two key applications:
Ultra-high fidelity amplification of bisulfite-converted, deaminated, or damaged DNA templates
Contamination control in PCR systems using dUTP and heat-labile UDG
Additionally, UniProof is widely used in USER cloning workflows.

One unit is defined as the amount of enzyme required to catalyze the incorporation of 10 nmol dNTP into acid-insoluble material within 30 minutes at 72°C.
Storage Conditions
- Storage buffer: 10 mM Tris-HCl (pH 7.6), 100 mM NaCl, 0.05% Tween-20, 1 mM DTT, 0.5 mM EDTA, 50% glycerol.
- Storage temperature: Stable at –20°C for up to 3 years.
