Description

5× UniProof HiFi PCR Mix is an optimized PCR master mix containing UniProof HiFi DNA Polymerase, dNTPs, Mg²⁺, a Mg²⁺-based heat-activation system, proprietary reaction buffer, and blue electrophoresis tracking dyes. Simply add primers and template to initiate high-performance PCR.

UniProof DNA Polymerase integrates the strengths of Q5 and Q5U enzymes. Engineered from a high-fidelity polymerase scaffold, it features an enhanced extension domain for robust, high-yield amplification of long fragments—up to 8 kb from genomic DNA and 20 kb from λDNA. The enzyme demonstrates an extension speed of 2 kb/min, with 5'→3' polymerase and strong 3'→5' exonuclease (proofreading) activity, achieving fidelity 280× that of Taq polymerase and producing blunt-ended products.

This mix employs a hot-start activation mechanism using Mg²⁺ sequestration for high specificity. Upon addition of Blocker15, a precipitate forms with Mg²⁺, fully inhibiting polymerase activity at ambient temperatures. During a 2-minute, 95°C pre-denaturation step, the Activator hydrolyzes the Blocker, releasing Mg²⁺ ions and activating the enzyme—initiating precise amplification with minimized non-specific products.

One unit is defined as the amount of enzyme required to catalyze the incorporation of 10 nmol dNTP into acid-insoluble material within 30 minutes at 72°C.

• Storage buffer: 10 mM Tris-HCl (pH 7.6), 100 mM NaCl, 0.05% Tween-20, 1 mM DTT, 0.5 mM EDTA, 50% glycerol.
