Description
Tma Endonuclease III is derived from a thermophilic bacterium and is a homologous protein to E. coli Endonuclease III, displaying excellent thermal stability. It possesses both N-glycosylase activity and AP-lyase activity. The N-glycosylase activity can release oxidatively damaged pyrimidine bases occurring on double-stranded DNA, generating an apurinic/apyrimidinic (AP) site. Subsequently, the AP-lyase activity cleaves at this site. Tma Endonuclease III recognizes DNA damage, including abasic sites on DNA, 5,6-dihydroxythymine, and thymine glycol. Our Tma Endonuclease III is recombinantly expressed and prepared through multi-step purification of the recombinant protein.
Activity Definition
One unit of activity is defined as the amount of enzyme required to cleave 1 pmole of a 60-mer oligonucleotide double-stranded DNA containing a single AP site in 1 hour at 65°C in a reaction buffer system of 1× Tma Endonuclease III.
Activity Assay Conditions
1× AM Buffer E: 20 mM Tris-HCl (pH 8.8), 10 mM KCl, 10 mM (NH4)2SO4, 2 mM MgSO4, 0.1% Triton X-100, incubation at 65°C.
