Description
SBS Matrix Gel Basement Membrane Matrix, also known as ECM (Extracellular Matrix) or BME (Basement Membrane Extract) gel, is free from Lactate Dehydrogenase Elevating Virus (LDEV). It is widely used in research areas such as angiogenesis, tumor cell migration, in vivo tumor model establishment, 3D tumor cell models, and organoids.
The matrix gel is extracted from the Engelbreth-Holm-Swarm (EHS) mouse tumor, which is rich in extracellular matrix proteins. Its main components include laminin, type IV collagen, heparan sulfate proteoglycans, entactin, various growth factors, tissue plasminogen activator, and trace amounts of matrix metalloproteinases. The entire product range is free from lactate dehydrogenase elevating virus or other viruses from mouse and tumor origins. At room temperature, the matrix gel can rapidly polymerize to form a biologically active 3D culture matrix, simulating the structure, composition, physical properties, and functions of the in vivo cell basement membrane. It promotes the adhesion, maintenance, growth, and differentiation of various cells, including endothelial cells, epithelial cells, hepatocytes, neurons, melanoma cells, thyroid cells, and follicular cells. It is used to study cell morphology, biochemical functions, migration, invasion, and gene expression.
The SBS Matrix Gel series includes various types of matrix gels with/without phenol red, including Standard, Growth Factor Reduced (GFR), High Concentration (HC), High Concentration Growth Factor Reduced (HC, GFR), hESC-Qualified for stem cells, and For Organoid Culture. SBS Matrix Gel without phenol red is suitable for fluorescence detection or colorimetric analysis; low growth factor SBS Matrix Gel is suitable for experiments requiring strict matrix composition, such as cell signaling pathways and cytokine studies; high concentration SBS Matrix Gel is suitable for angiogenesis research, tumor cell migration, and in vivo tumor model establishment. Features
- The composition of this matrix gel is similar to that of conserved, early developmental basement membranes. The primary components of the matrix gel are approximately 60% laminin, 30% type IV collagen, and 8% entactin. Entactin is a bridging molecule that interacts with laminin and type IV collagen, contributing to the structural organization of extracellular matrix molecules. Besides the main components, the matrix gel also contains proteoglycans and growth factors such as perlecan, vascular endothelial growth factor (VEGF), transforming growth factor-β (TGF-β), epidermal growth factor (EGF), insulin-like growth factor (IGF), fibroblast growth factor (FGF), tissue plasminogen activator (PLAT), and naturally occurring growth factors in tumors. Additionally, there are residual matrix metalloproteinases derived from tumor cells.
- The matrix gel rapidly polymerizes at room temperature. It begins to gel at temperatures above 10ºC. At 22-37ºC, the macromolecules in the matrix gel form chemical bonds, prompting rapid polymerization to form a 3D gel matrix. However, at lower temperatures (e.g., 4ºC), insufficient energy prevents chemical bonding, keeping the matrix gel in a liquid state.
- This matrix gel meets different experimental needs. The higher the protein concentration in the matrix gel, the higher the viscosity. The protein concentration of non-high concentration matrix gel is 8-13mg/ml, while high concentration matrix gel has a protein concentration of 16-26mg/ml. The minimum gelling concentration of the matrix gel is 3mg/ml. To avoid incomplete gelling, the final concentration of matrix gel for in vivo studies should not be less than 4mg/ml.
- The matrix gel undergoes strict quality control, ensuring high quality. Each batch of related products is rigorously tested for protein concentration, endotoxins, gel formation, angiogenesis, 3D tumor cell culture, organoid culture, biological activity, sterility (bacteria, fungi, viruses, mycoplasma), and more.
Storage
Store at -20℃ or below, protected from light, for two years. After the initial thaw, aliquot appropriately to avoid repeated freeze-thaw cycles.
Precautions
- This product is shipped on dry ice and is solid before thawing. If there is no dry ice in the foam box upon receipt and the matrix gel appears gel-like instead of liquid, please contact us immediately. If you do not aliquot or directly use it upon receipt, store it at -20℃ or below, protected from light.
- The matrix gel cannot be repeatedly freeze-thawed. To ensure optimal use, aliquot the product based on single-use amounts upon receipt, minimizing freeze-thaw cycles, or directly purchase the 1ml small packaging.
- Store the matrix gel at -20℃ or below, protected from light, and maintain it in a frozen state during storage. Due to the automatic defrosting feature of frost-free refrigerators, which may affect the internal temperature, it is not recommended to store this product in frost-free refrigerators.
- The protein concentration of the matrix gel varies between batches and is generally indicated on the vial label. Determine the usage amount based on the indicated protein concentration and the required concentration to obtain consistent results. Optimize the dilution concentration according to the experiment.
- Use aseptic techniques to avoid contamination when handling this product. After thawing, place the product in a sterile area and spray the top with 70% ethanol for disinfection and air dry.
- Thawed matrix gel can be kept liquid for about 30 days if stored in the dark at 4℃ or on ice, but it is still recommended to aliquot or use it for related experiments immediately after thawing.
- Due to the effects of carbon dioxide and bicarbonate buffer and phenol red (if present), the matrix gel may change color during freeze-thaw cycles. This color change is normal and does not affect product performance.
- When handling the matrix gel during experiments, avoid touching the gel area of the vial with your hand to prevent gelation caused by body heat. Hold the vial/tube above the gel area.
- Do not save leftover matrix gel for future use after completing the experiment.
- This product is intended for scientific research use by professionals only and is not for clinical diagnosis or treatment, food, or drugs. Do not store in residential areas.
- For your safety and health, wear a lab coat and disposable gloves while handling.
